摘要
旨在研究微贮豆秸中添加维生素D_(3)(VD_(3))对绵羊组织的抗氧化性能及细胞凋亡的影响。随机选取25只2月龄健康绵羊,分为5组,对照组日粮中不添加VD_(3),试验组日粮中分别添加300,600,900,1200 IU/kg的VD_(3),连续饲喂80 d。结果显示:(1)饲粮中添加VD_(3)后,绵羊血清、肝脏、脾脏组织的T-SOD活性显著升高(P<0.05),胸腺GSH-Px活性极显著升高(P<0.01),血清、肝脏、脾脏、胸腺及肠系膜淋巴结组织的MDA含量显著降低(P<0.05或P<0.01);添加300,600,900 IU/kg VD_(3)时,血清GSH-Px活性显著升高(P<0.05);添加300,900,1200 IU/kg VD_(3)后,肾脏T-SOD活性显著升高(P<0.05);添加300,600 IU/kg VD_(3)时,肾脏MDA含量显著降低(P<0.05);添加300,900 IU/kg VD_(3)时,肠系膜淋巴结T-SOD活性显著升高(P<0.05或P<0.01)。(2)饲粮添加300 IU/kg VD_(3)时,脾脏和肾脏组织的Caspase-9、VDR和BAX含量显著降低(P<0.05),肝脏、脾脏、胸腺和肠系膜淋巴结组织的Bcl-2表达量显著升高(P<0.05);添加600 IU/kg VD_(3),肝脏和肾脏组织的Caspase-3、Caspase-9、VDR和BAX表达量显著升高(P<0.05);添加900,1200 IU/kg VD_(3),肝脏,胸腺和肠系膜淋巴结组织的Caspase-3表达量显著升高(P<0.05)。以上结果表明,饲粮中适当添加VD_(3)可有效改善绵羊的抗氧化性能,调节凋亡相关基因的表达,抑制细胞凋亡。本试验条件下,添加300 IU/kg VD_(3)可显著改善绵羊组织的抗过氧化能力,减缓各组织细胞的凋亡。
This study aims to study the effect of VD_(3)added in micro-storage bean straw on the anti-oxidation performance and cell apoptosis of sheep tissues.Twenty-five 2-month-old healthy sheep were randomly selected and divided into five groups.The diet of the control group was not supplemented with VD_(3),and the diet of the test group was supplemented with 300,600,900,1200 IU/kg of VD_(3),consecutively.The experiment lasts for 80 d.The results show that,(1)after the addition of VD_(3)to the diet,T-SOD activity of sheep serum,liver and spleen tissues increased significantly(P<0.05);GSH-Px activity of thymus tissue extremely significantly increased(P<0.01);the MDA content in serum,liver,spleen,thymus and mesenteric lymph nodes was significantly reduced(P<0.05 or P<0.01);when 300,600,900 IU/kg VD_(3)were added,the serum GSH-Px activity was significantly increased(P<0.05);when 300,900,1200 IU/kg VD_(3)were added,T-SOD activity of kidney tissue was significantly increased(P<0.05),when 300,600IU/kg VD3 were added,MDA content in kidney tissue was significantly reduced(P<0.05);when 300,900IU/kg VD3 were added,the T-SOD activity of mesenteric lymph nodes was significantly increased(P<0.05or P<0.01).(2)When 300IU/kg VD3 was added to the diet,the contents of Caspase-9,VDR and BAX in the spleen and kidney tissues were significantly reduced(P<0.05),and the expression of Bcl-2in the liver,thymus and mesenteric lymph nodes tissues was significantly increased(P<0.05);with the addition of 600IU/kg VD3,the expression of Caspase-9,VDR and BAX in spleen and kidney tissues increased significantly(P<0.05);with the addition of 900,1200IU/kg VD3,the expression of Caspase-3in kidney,spleen,thymus and mesenteric lymph nodes tissues increased significantly(P<0.05).It is shown that the appropriate addition of VD3to the diet can effectively activate apoptosis-related genes,and induce cell apoptosis.When the dosage is 300IU/kg,the anti-peroxidation ability of sheep tissue can be significantly improved,and the apoptosis of various tissue cells is
