摘要
本试验旨在研究日粮中添加甘氨酸(Gly)对脂多糖(LPS)应激断奶仔猪肠黏膜屏障损伤的保护作用。选用24头21日龄断奶仔猪,随机分为4组,即对照组(基础日粮)、LPS组(基础日粮+LPS)、1.0%Gly组(基础日粮+1.0%Gly+LPS)、2.0%Gly组(基础日粮+2.0%Gly+LPS)。预饲期7 d,正试期28 d,在试验第28天,LPS组、1.0%Gly组和2.0%Gly组仔猪腹腔注射LPS(100μg/kg BW),对照组注射无菌生理盐水。注射4 h后,仔猪屠宰,取脾脏、肝脏、肠系膜淋巴结及空肠、回肠样品待测。结果表明:①各处理组的平均日增重、平均日采食量及耗料增重比无显著差异。②与对照组相比,LPS应激升高了回肠丙二醛(MDA)水平(P<0.05),降低空肠、回肠谷胱甘肽(GSH)水平(P<0.05);与LPS组相比,日粮中添加Gly降低回肠MDA水平、升高GSH水平(P<0.05)。③与对照组相比,LPS应激增加肝脏移位细菌数(P<0.05);与LPS组相比,日粮中添加Gly降低肝脏移位细菌数(P<0.05)。④与对照组相比,LPS应激降低空肠绒毛高度/隐窝深度比值(P<0.05),且有降低空肠绒毛高度趋势(P<0.1);与LPS组相比,日粮中添加Gly对空肠、回肠的绒毛高度、隐窝深度及绒毛高度/隐窝深度比值没有显著影响。⑤与对照组相比,LPS应激增加回肠肥大细胞密度(P<0.05);与LPS组相比,日粮中添加Gly降低空肠肥大细胞密度(P<0.05)。⑥与对照组相比,LPS应激升高空肠、回肠CRF(P<0.05)、CRFR1(P<0.001)和GR(P<0.05)的mRNA相对表达量;与LPS相比,日粮添加Gly降低了空肠、回肠CRFR1(P<0.05)及空肠GR(P<0.001)的mRNA相对表达量。结果提示,日粮添加Gly可缓解LPS应激导致的肠黏膜屏障功能障碍。
The aim of this study was to investigate the protective effect of glycine(Gly)on injury of intestinal mucosal barrier induced by lipopolysccharide(LPS)in weaned pigs.Twenty-four pigs weaned at 21 d of age were randomly divided into four groups:control group(basal diet),LPS group(basal diet+LPS),1.0%Gly group(basal diet+1.0%Gly+LPS),and 2.0%Gly group(basal diet+2.0%Gly+LPS).Pigs were acclimated for 7 days to experimental conditions and fed control diet.The experiment lasted for 28 days.On d 28 of the trial,pigs were injected intraperitoneally with saline or LPS.After 4 hours of LPS or saline injection,pigs were slaughtered and spleen,mesenteric lymph,liver and intestine were collected.The results showed that:①There was no difference in average daily gain(ADG),average daily feed intake(ADFI)and feed/gain(F/G)among four treatments(P>0.05).②Compared with control group,LPS challenge increased the level of MDA in ileum(P<0.05),and decreased the level of GSH in jejunum and ileum(P<0.05).Compared with LPS group,Gly supplementation decreased the level of MDA and increased the level of GSH in ileum(P<0.05).③Compared with control group,LPS challenge increased bacterial translocation incidence and translocation microorganisms in the spleen(P<0.05).Compared with LPS group,Gly supplementation reduced translocation microorganisms in the liver(P<0.05).④Compared with the control group,LPS challenge decreased jejunal villus height/crypt depth ratio(P<0.05),and tended to decrease jejunal villus height(P<0.1).Compared with LPS group,Gly supplementation had no effects on villus height,crypt depth and villus height/crypt depth ratio in jejunum and ileum(P>0.05).⑤Compared with control group,LPS challenge increased mast cell density in ileum(P<0.05).Compared with LPS group,Gly supplementation decreased the density of jejunal mast cells(P<0.05).⑥Compared with control group,LPS challenge increased the mRNA expression of CRF(P<0.05),CRFR1(P<0.001),GR(P<0.05)in jejunum and ileum.Compared with LPS group,Gly supplementation decrea
