丹参多糖缓解小鼠乳腺炎症和过氧化反应的作用与机制 被引量:3

Effect and mechanism of Salvia miltiorrhiza polysaccharides in alleviating mammary gland inflammation and peroxidation in mice

摘要 为探究丹参多糖对乳腺炎症损伤的缓解作用和机理,将60只分娩昆明母鼠分为空白对照组、模型组和丹参多糖用药组(250,500,750 mg/kg),从分娩结束开始记作第1日,连续灌胃7 d,第8日使用LPS建立乳腺炎模型,24 h后眼球取血并剥离保存乳腺组织。观察比较小鼠临床表现和乳腺外观变化;通过H&E染色比较小鼠乳腺病理组织学变化;通过ELISA测定血清中IL-1β、IL-6和TNF-α水平;通过生化试剂盒检测乳腺组织总一氧化氮合成酶(T-NOS)、还原型谷胱甘肽(GSH)、丙二醛(MDA)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)水平;采用IHC法半定量分析IκB-α、IKK和NF-κB表达水平;通过qPCR检测IκB-α、IKK和NF-κB的mRNA表达水平。结果表明,与空白对照组小鼠相比,模型组小鼠乳腺组织出现明显红肿和出血点,镜下可见大量炎性细胞浸润;丹参多糖用药组较模型组T-NOS活力和MDA含量显著降低(P<0.05或P<0.01),SOD、CAT活力和GSH水平显著升高(P<0.05或P<0.01);丹参多糖用药组NF-κB通路相关蛋白表达水平均显著低于模型组(P<0.05或P<0.01);IκB-α、IKK和NF-κB在mRNA转录水平上,除丹参多糖低剂量组的NF-κB mRNA水平与模型组无差异外(P>0.05),各剂量组的其余指标与模型组相比有显著降低(P<0.05或P<0.01)。以上结果表明丹参多糖可显著缓解小鼠乳腺组织炎症和过氧化反应,其作用途径可能是通过抑制NF-κB信号通路的激活改善炎症损伤。本研究为临床上应用丹参多糖防治奶牛乳腺炎提供了理论依据。 The purpose of the present study was to investigate the effects of Salvia miltiorrhiza polysaccharides(SMPs)on LPS-induced mastitis.Sixty delivery female mice were divided into the blank control group,model group and three different doses of SMPs groups(250,500,750 mg/kg),SMPs were given by intragastric administration for one week before modeling.The mouse model of mastitis was induced by inoculation of LPS through the canals of the mammary gland.The mammary gland and blood were collected 24 h after induction.The changes of clinical manifestation and mammary gland appearance were observed and compared in different groups.Histopathological changes of mammary gland were observed by H&E staining.The levels of IL-1β,IL-6,TNF-α,total nitric oxide synthase(T-NOS),malondialdehyde(MDA),glutathione(GSH),superoxide dismutase(SOD)and catalase(CAT)in mammary gland were detected by biochemical kits.The protein expression level of IκB-α,IKK and NF-κB were detected by semi-quantitative immunohistochemistry(IHC)analysis.The results showed that compared with the blank control group,the mammary glands of the model group had obvious redness and bleeding spots.Numerous inflammatory cell infiltrates were observed microscopically.Compared with model group,the activities of T-NOS and MDA content were significantly decreased(P<0.05or P<0.01),and the activities of SOD,CAT and GSH were significantly increased in SMPs treatment groups(P<0.05or P<0.01).The expression level of NF-κB pathway related proteins in the SMPs treatment groups was significantly lower than that in the model group(P<0.05or P<0.01).In terms of mRNA expression levels of IκB-α,IKK and NF-κB,except for the low-dose group,which the NF-κB mRNA level was not statistically different from the model group(P>0.05),the other indexes in each dose group were significantly lower than those in the model group(P<0.05or P<0.01).These results suggested that SMPs could significantly alleviate the inflammation and peroxidation in mouse mammary glands,which might be mediated by i
