内关穴位埋线对大鼠心肌梗死后心肌肥大的影响及作用机制 被引量:3

Effect and Mechanism of Acupoint Catgut Embedding at Neiguan on Myocardial Hypertrophy after Myocardial Infarction in Rats

摘要 目的 观察内关埋线对大鼠心肌梗死后心肌肥大的影响,探究其作用机制。方法 采取结扎大鼠左冠状动脉前降支的方法建立心肌梗死动物模型,将其随机分为模型组、内关埋线组、培哚普利组,另设假手术组,其中假手术组只穿线不结扎,每组5只,术后第2天开始干预治疗,干预治疗7 d后,检测大鼠心脏功能,并观察心脏梗死边缘区心肌细胞形态学改变,检测心肌梗死边缘区ANP、m TOR、p-P70S6K、P70S6K、p-4E-BP1、4EBP1的蛋白表达情况。结果 1)与假手术比较,模型组大鼠左室射血分数(LVEF)、左室短轴率(LVFS)明显降低,左室舒张末期内径(LVIDd)、左室收缩末期内径(LVIDs)显著升高,与模型组比较,内关埋线组和培哚普利组LVEF、LVFS显著升高,LVIDd、LVIDs显著降低(P <0.05)。2)与假手术组比较,模型组横截面积显著升高,与模型组比较,内关埋线组和培哚普利组横截面积显著降低(P <0.05)。3)与假手术组比较,模型组ANP蛋白表达显著升高,与模型组比较,内关埋线组和培哚普利组ANP蛋白表达降低(P <0.05)。4)与假手术组比较,模型组p-mTOR/mTOR、p-P70S6K/P70S6K、p-4EBP1/4EBP1蛋白表达水平显著升高(P <0.05);与模型组比较,内关埋线组、培哚普利组p-mTOR/mTOR、p-P70S6K/P70S6K、p-4EBP1/4EBP1蛋白表达水平显著降低(P <0.05)。结论 内关埋线能够改善大鼠心肌梗死后心肌肥大的程度,其机制可能与抑制mTOR/p70S6K/4E-BP1信号通路有关。 Objective:To observe the effect of Neiguan catgut embedding on myocardial hypertrophy after myocardial infarction in rats and to explore its mechanism. Methods:Myocardial infarction animal model was established by ligating the left anterior descending coronary artery in rats,and they were randomly divided into the model group,Neiguan catgut embedding group,perindopril group,and a sham operation group,in which the sham operation group only had threading without ligation. There were 5 rats in each group. The intervention treatment was started on the second day after the operation. After 7 days of intervention treatment,the cardiac function of the rats was detected,and the morphological changes of myocardial cells in the marginal area of myocardial infarction were observed,and ANP,m TOR,p-P70S6K,P70S6K,p-4E-BP1,4E-BP1 were detected. Results:1)Compared with sham operation group,the left ventricular ejection fraction and left ventricular short axis rate in the model group were significantly decreased,and the left ventricular end-diastolic diameter and left ventricular end-systolic diameter were significantly increased. Compared with the model group,the LVEF and LVFS in the internal fixation group and perindopril group were significantly increased,and the LVIDd and LVIDs were significantly decreased(P < 0.05). 2)Compared with the sham operation group,the cross-sectional area of the model group was significantly increased,and compared with the model group,the cross-sectional areas of the internal fixation group and the perindopril group were significantly decreased(P < 0.05). 3)Compared with the sham operation group,the expression of ANP protein in the model group was significantly increased. Compared with the model group,the ex-pressions of ANP protein in the Neiguan catgut embedding group and the perindopril group were decreased(P <0.05). 4)Compared with the sham operation group,the protein expression levels of p-mTOR/mTOR,p-P70S6K/P70S6K and p-4EBP1/4EBP1 in the model group were significantly increased(P < 0.05).
