摘要
以猪皮中提取、水解得到的I型胶原和明胶为研究对象,研究了二者对羟基自由基的清除影响。紫外光谱结果表明,胶原多肽和明胶均对羟基自由基具有清除能力,二者清除羟基自由基的能力与其浓度呈正相关,而且明胶清除自由基的能力明显优于胶原多肽。氨基酸分析结果表明,I型胶原和明胶分子中氨基酸种类及含量相近,这不是造成二者清除自由基能力差异的关键。圆二色谱分析结果表明,明胶分子包含α-螺旋、β-折叠、β-回转、任意卷曲四种二级结构,其中β-折叠、β-回转、任意卷曲结构含量总和远远高于α-螺旋,表明明胶分子链处于较高伸展度,有利于抗氧化氨基酸暴露,有利于清除自由基。而I型胶原蛋白的三条肽链聚合在一起,其中的氨基酸基团暴露的相对较少,其溶液中游离的羟基自由基有很小机会可以和这些氨基酸基团结合,限制了自由基的清除能力。因此,二者结构上的不同造成了明胶的羟基自由基的清除率高于胶原多肽。本研究将为胶原蛋白抗氧化的研究提供理论和应用基础。
In this paper,type I collagen and gelatin extracted and hydrolyzed from pig skin were used to study their effects on hydroxyl radical scavenging.The results of UV spectra showed that both collagen polypeptide and gelatin had the ability to scavenge hydroxyl radicals,and their ability to scavenge hydroxyl radicals was positively correlated with their concentration,and the ability of gelatin to scavenge hydroxyl radicals was significantly better than that of collagen polypeptide.The results of amino acid analysis showed that the type and content of amino acids in type I collagen and gelatin were similar,which was not the key to the difference of free radical scavenging ability between them.Circular dichroism analysis showed that gelatin molecules containedα-Spiralβ-Collapseβ-Rotary,arbitrary curl four secondary structures,whereinβ-fold,β-The total content of rotation and arbitrary curl structure is much higher thanα-Helix,indicating that the gelatin molecular chain is in a high degree of extension,which is conducive to the exposure of antioxidant amino acids and the elimination of free radicals.The three peptide chains of type I collagen are polymerized together,in which the amino acid groups are relatively less exposed,and the free hydroxyl radicals in the solution have little chance to combine with these amino acid groups,which limits the scavenging ability of free radicals.Therefore,the difference in the structure of gelatin and collagen resulted in higher hydroxyl radical scavenging rate than collagen peptides.This study will provide a theoretical and applied basis for the study of collagen antioxidation.
