摘要
目的:探索云南道地药材滇黄精中黄精多糖抗糖尿病的作用。方法:采用水提醇沉法提取滇黄精总多糖,分级醇沉法分离出均一多糖,再经DEAE-FF琼脂糖凝胶柱纯化。SPF级ICR小鼠雄性75只,随机选出15只作为空白对照组,正常饮食喂养。剩余采用高脂高糖饮食联合低剂量链脲佐菌素制备2型糖尿病小鼠模型,造模成功后,分为4组:即模型组,阳性组、滇黄精总多糖组(PPS)、滇黄精均一多糖组(PPS1)。观察各组小鼠的空腹血糖(FBG)、血脂水平、肝原含量、肌糖原含量,以及肾脏、胰腺病理改变情况。结果:滇黄精总多糖和滇黄精均一多糖能显著降低糖尿病小鼠的空腹血糖值,降低TC、TG、LDL水平,且均能减少糖尿病小鼠肾脏和胰腺的破坏。其中,滇黄精总多糖可明显增加糖尿病小鼠肝糖原和肌糖原含量,亦可显著升高糖尿病小鼠HDL水平。结论:滇黄精总多糖和滇黄精均一多糖均有一定抗2型糖尿病的作用,但两者作用效果存在差异。
Objective To explore the mechanism of the anti-diabetes of Polygonatum kingianum polysaccharide by pharmacodynamics study.Methods PPS was fractioned from Polygonatum kingianum by ethanol subsiding method,PPS1 are usde by fractional alcohol precipitation method,and purified by DEAE-FFCellulose chromatography.The high fat and high glucose diet combined with low dose streptozotocin were applied in the 60 SPF-level male mice prepare T2DM rats models.They were randomly divided into model group,positive group,PPS group and PPS1 group.Another 15 mice were enrolled as control group.The levels of fasting blood glucose(FBG),cholesterol(TC),triacylglycerol(TG),muscle glycogen and liver glycogen were observed.Results PPS and PPS1 essence can decrease the fasting blood glucose value of diabetic mice,decrease TC,TG and LDL level,and can reduce the damage of kidney and pancreas in diabetic mice.Conclusion PPS and PPS1 have obvious activity anti-diabetes.
