摘要
目的 探讨高良姜素(Galangin,GLA)对人卵巢癌SKOV3细胞增殖、迁移和凋亡能力的影响。方法 用不同浓度的GLA(0μmol/mL、10μmol/mL、20μmol/mL、40μmol/mL)处理卵巢癌SKOV3细胞24 h,通过CCK8法和划痕实验来检测GLA对SKOV3细胞增殖和迁移的影响;通过TUNEL实验和流式细胞术检测GLA对SKOV3细胞凋亡的影响;通过Western Blot实验检测B细胞淋巴瘤-2蛋白(B-cell lymphoma-2,Bcl-2)、Bcl-2-Associated X蛋白(Bcl-2-Associated X,Bax)、蛋白激酶B(protein kinase B,AKT)和磷酸化蛋白激酶B(phosphor-protein kinase B,p-AKT)蛋白表达水平。结果 CCK8和划痕实验的结果显示,GLA能够抑制卵巢癌SKOV3细胞的增殖和迁移能力(P<0.05),且呈剂量依赖性;TUNEL实验和流式细胞术检测结果显示,GLA以剂量依赖的方式诱导了卵巢癌细胞的凋亡(P<0.05);Western Blot实验结果显示GLA抑制了Bcl-2蛋白表达,增强了Bax蛋白表达,并且抑制了PI3K/Akt信号通路中p-AKT蛋白的表达(P<0.05)。结论 GLA可能通过PI3K/AKT信号通路抑制了卵巢癌SKOV3细胞增殖和迁移能力,诱导了细胞的凋亡。
Objective To investigate the effects of galangin(GLA) on proliferation, migration and apoptosis of human ovarian cancer SKOV3 cells. Methods With different concentrations of GLA(0 μmol/mL, 10 μmol/mL, 20 μmol/mL, 40 μmol/mL) for 24 hours, and the effects of GLA on the proliferation and migration of SKOV3 cells were detected by CCK8 method and scratch test;the effect of GLA on SKOV3 cell apoptosis was detected by TUNEL assay and flow cytometry;the protein expression levels of B-cell lymphoma-2(Bcl-2), Bcl-2-associated x(Bax), protein kinase B(AKT) and phosphor protein kinase B(p-AKT) were detected by Western Blot. Results The results of CCK8 and scratch test showed that GLA could inhibit the proliferation and migration of ovarian cancer SKOV3 cells in a dose-dependent manner(P<0.05);TUNEL test and flow cytometry showed that GLA induced apoptosis of ovarian cancer cells in a dose-dependent manner(P<0.05);Western blot showed that GLA inhibited the expression of Bcl-2 protein, enhanced the expression of Bax protein and inhibited the expression of p-AKT protein in PI3 K/AKT signal pathway(P<0.05).Conclusion GLA inhibits the proliferation and migration of ovarian cancer SKOV3 cells through PI3 K/AKT signaling pathway and induces cell apoptosis.
