雌二醇和孕酮对脂多糖诱导的去卵巢小鼠输卵管组织中IL-1βmRNA表达的影响

Effects of Estradiol and Progesterone on IL-1β mRNA Expression in Oviduct Tissue of LPS-induced Ovariectomized Mice

摘要 目的研究雌二醇(E2)和孕酮(P4)对脂多糖(LPS)诱导的去卵巢小鼠急性输卵管炎症组织中白细胞介素-1β(IL-1β)mRNA表达的影响,初步探讨雌、孕激素的抗炎机制。方法将健康雌性KM小鼠随机分为假手术组(LPS+SHAM)、卵巢摘除组(LPS+OVX),卵巢摘除+雌二醇组(LPS+OVX+E2)、卵巢摘除+孕酮组(LPS+OVX+P4)、LPS组及对照组(control),应用HE染色观察各组小鼠输卵管组织病理改变、RT-qPCR法检测各组小鼠输卵管中IL-1βmRNA表达水平。结果观察输卵管组织病理学改变,雌、孕激素可明显减轻LPS所致的输卵管炎症;与对照组比较,LPS组、LPS+SHAM组和LPS+OVX+E2组IL-1βmRNA表达显著下调,LPS+OVX+P4组IL-1βmRNA表达极显著下调;与LPS+OVX组比较,LPS+OVX+E2组IL-1βmRNA表达显著下调,LPS+OVX+P4组IL-1βmRNA表达极显著下调。结论雌、孕激素可抑制小鼠输卵管炎症组织内IL-1βmRNA的表达,从而起到减轻LPS所致的炎症反应的作用。 Objective To study the effect of estradiol(E2)and progesterone(P4)on the expression of interleukin-1β(IL-1β)mRNA in lipopolysaccharide(LPS)-induced acute fallopian tube inflammation in ovariectomized mice,and preliminarily explore the anti-inflammatory mechanism of estrogen and progesterone.Methods Healthy female KM mice were randomly divided into sham operation group(LPS+SHAM),ovarian removal group(LPS+OVX),ovarian removal+estradiol group(LPS+OVX+E2),ovarian removal+progesterone group(LPS+OVX+P4),LPS group and control group(control),HE staining was used to observe the pathological changes of the oviduct of each group of mice,and the expression level of IL-1βmRNA in the oviduct of each group of mice was detected by RT-qPCR.Results The histopathological changes of fallopian tube were observed.Estrogen and progesterone could significantly reduce the inflammation of fallopian tube caused by LPS.Compared with the control group,the expression of IL-1βmRNA in the LPS group,LPS+SHAM group and LPS+OVX+E2 group was significantly down-regulated,and the expression of IL-1βmRNA in the LPS+OVX+P4 group was extremely significantly down-regulated;Compared with the LPS+OVX group,the expression of IL-1βmRNA in the LPS+OVX+E2 group was significantly down-regulated,and the expression of IL-1βmRNA in the LPS+OVX+P4 group was extremely significantly down-regulated.Conclusion Estrogen and progesterone can inhibit the expression of IL-1βmRNA in the inflammatory tissue of the oviduct of mice,thereby reducing the inflammatory response caused by LPS.
