摘要
几丁质酶是一种病程相关蛋白,不但在植物对病原菌和虫害的防御过程中发挥重要作用,也参与了植物对非生物胁迫的应答反应。然而,目前对几丁质酶在水稻应答各种生物和非生物胁迫的相关研究报道相对较少。利用BLASTP和隐马科夫模型(HMM),在水稻基因组中鉴定了49个几丁质酶基因,其中33个几丁质酶基因属于GH18亚家族,16个基因属于GH19亚家族,位于同一亚族的几丁质酶基因结构和保守基序组成相似。染色体定位分析表明,除了12号染色体以外,水稻几丁质酶基因不均匀地分布在剩下的11条染色体上。53.06%(26/49)的水稻几丁质酶基因发生了复制,其中包括4对片段重复基因和18个串联重复基因。通过RNA-seq数据分析水稻几丁质酶家族基因在不同器官/组织中的表达模式,发现水稻几丁质酶家族基因具有明显的组织特异性表达模式。此外,大部分的几丁质酶基因在叶和/或根中的表达至少受到一种胁迫因子的诱导。总的来说,水稻几丁质酶基因家族的综合分析将有助于进一步了解几丁质酶基因在水稻生长发育中的作用。
Chitinases,a subgroup of pathogenesis-related proteins,play a key role in plant defense against pathogens and pests and are also involved in abiotic stress response.However,the functions of chitinases involved in biotic and abiotic stresses of rice(Oryza sativa L.)remain largely unreported.In the present study,a total of 49 putative chitinase genes were identified in the rice genome through BLASTP and HMM(Hidden Markov Model),of which 33 chitinases genes belong to the GH18 subfamily,16 genes belong to the GH19 subfamily,and the same subfamily shared similar conserved motif composition and gene structure.Chromosomal location analysis indicated that chitinases genes were unevenly distributed in 11 of 12rice chromosomes.In total,53.06%(26/49)of the chitinases genes were expanded via gene duplication,18 and 8 rice chitinases genes were tandemly and segmentally duplicated,respectively.Comprehensive analyses were performed to investigate the expression profiles of rice chitinases genes in various tissues/organs at different development stages by using data from RNA-seq result.Many chitinases genes in rice exhibited abundant and tissue-specific expression patterns.Moreover,most chitinases genes were found to be differentially expressed at least one stressor in leaves and/or roots.Taken together,our study provides a comprehensive analysis of the chitinases gene family and will facilitate further studies on their roles in rice growth and development.
