番泻叶总黄酮的提取工艺优化及体外抗氧化活性研究 被引量:2

摘要 在单因素实验的基础上,采用响应面法优化番泻叶总黄酮的提取工艺,通过测定其对DPPH自由基的清除率来评价其体外抗氧化活性,并对抗氧化实验进行方法学考察。结果表明,番泻叶总黄酮的最佳提取工艺为:超声时间50 min、超声温度68℃、料液比1∶28(g∶mL),在此条件下,番泻叶总黄酮提取率为3.94%,RSD值为0.54%(n=3),与理论值相差0.05%;番泻叶总黄酮具有较强的体外抗氧化活性,当其浓度为0.039 mg·mL^(-1)时,其对DPPH自由基的清除率为60.5%。 We optimized the extraction process of total flavonoids from Cassia angustifolia leaves by response surface methodologies on the basis of single-factor experiments.Moreover,we evaluated their antioxidant activity in vitro by the determination of the scavenging rate of DPPH free radicals,and investigated the methodology of antioxidation experiments.The results show that the optimal extraction conditions are determined as follows:the ultrasonic time of 50 min,the ultrasonic temperature of 68℃,and the solid-liquid ratio of 1∶28(g∶mL).Under above conditions,the extraction rate of total flavonoids from Cassia angustifolia leaves can reach 3.94%,and the RSD value is 0.54%(n=3),which differs 0.05%from the theoretical value.When the concentration of total flavonoids from Cassia angustifolia leaves is 0.039 mg·mL^(-1),the scavenging rate of DPPH free radicals can reach 60.5%,indicating that the total flavonoids from Cassia angustifolia leaves have strong antioxidant activity in vitro.
