摘要
以二氢杨梅素(DMY)为底物、Na_(2)SeO_(3)为硒化剂制备了硒化DMY。采用UV-Vis、FTIR、NMR、XRD、TG、原子荧光光谱表征其结构和性能。用CCK-8法检测了硒化DMY对人口腔鳞癌细胞HSC-3细胞增殖的影响,通过划痕实验研究了硒化DMY对HSC-3细胞迁移的影响。结果表明,硒化DMY中仍存在黄酮基本母核,并新形成C—Se键,其中硒含量为6.54%±0.22%;DMY和硒化DMY均对HSC-3细胞的增殖和迁移有良好的抑制作用,抑制效果与其质量浓度呈正相关性,且DMY和硒化DMY对HSC-3细胞的半数抑制质量浓度(IC_(50))分别为25.27、21.27μg/mL,DMY的硒化有效提高了其对HSC-3细胞增殖和迁移的抑制能力。
Selenizing dihydromyricetin(DMY)was prepared with DMY as substrate and Na_(2)SeO_(3)as selenizing agent.Its structure and performance were characterized by UV-Vis,FTIR,NMR,XRD,TG and atomic fluorescence spectrometry.The effects of selenizing DMY on human oral squamous cell carcinoma(HSC-3 cell)proliferation and human oral squamous cell carcinoma(HSC-3 cell)migration were tested by CCK-8 method and scratch experiment,respectively.The results showed that there was still a basic nucleus of flavonoids in selenizing DMY and new C—Se bonds were formed,and the selenium content was 6.54%±0.22%.Both DMY and selenizing DMY had a good inhibitory effect on the proliferation and migration of HSC-3 cells,and the inhibitory effect was positively correlated with the concentration.The half inhibitory mass concentration(IC_(50))of DMY and selenizing DMY on HSC-3 cells were 25.27 and 21.27μg/mL,respectively.The selenization of DMY effectively improved the ability to inhibit the proliferation and migration of HSC-3 cells.
