白藜芦醇调控线粒体通路诱导口腔鳞状细胞癌细胞凋亡 被引量:5

摘要 目的:研究白藜芦醇诱导人口腔鳞状细胞癌细胞(HN4、Cal27、HN6)凋亡的机制。方法:不同浓度白藜芦醇处理3种细胞后,通过MTT法检测细胞活力的变化;蛋白质免疫印迹法(Western blot)、实时荧光定量PCR分别从蛋白以及mRNA水平检测细胞中的凋亡相关因子以及线粒体通路关键因子表达量的变化。结果:MTT结果表明50~400μmol/L白藜芦醇对3种细胞活力均有抑制作用(P<0.05),且在50~400μmol/L浓度处理48 h后,3种细胞活力均低于24 h(P<0.05)。与对照组0μmol/L相比,在100、200μmol/L浓度处理48 h后,Bcl-2蛋白和mRNA的表达均降低(P<0.05);Bax蛋白和mRNA的表达均升高(P<0.05);cleaved-caspase3蛋白表达也升高(P<0.05);而cyt-c蛋白和mRNA的表达在50~200μmol/L浓度时升高(P<0.05)。结论:白藜芦醇可以诱导口腔鳞状细胞癌细胞凋亡,可能与调控凋亡相关蛋白Bcl-2、Bax的表达以及激活线粒体凋亡通路有关。 Objective:To study the mechanism of resveratrol in inducing apoptosis of human oral squamous cell carcinoma cells(HN4,Cal27,HN6).Methods:Human oral squamous cell carcinoma cells,including HN4,Cal27 and HN6,were initially treated with diverse dose of resveratrol,and then examined for their changes of viability using MTT method.Western blot and real-time quantitative PCR were performed to detect the changes apoptosis-related factor expression and expression quantity of the key factors in mitochondrial pathway at protein and mRNA levels.Results:MTT assay showed that resveratrol significantly inhibited viability of the three cells in dose of 50-400μmol/L(P<0.05),and the cellular viability was less than 24 h after treatment by concentration of 50-400μmol/L for 48 h(P<0.05).Compared with the control group at dose of 0μmol/L,the expression of both Bcl-2 protein and mRNA was significantly lower,yet Bax protein and mRNA expression and cleaved-caspase3 protein were markedly higher after treatment at concentration of 100 and 200μmol/L for 48 h(all P<0.05).In contrast,cyt-c protein and mRNA expression were significantly elevated at dose of 50-200μmol/L(P<0.05).Conclusion:Resveratrol can induce apoptosis of oral squamous carcinoma cells.The mechanism is potentially involved in regulation of the expression of apoptosis-related proteins Bcl-2 and Bax and activation of the mitochondrial apoptotic pathway.
