常用蛋白原料体外蛋白消化速率、肽含量和功能的研究 被引量:2

摘要 试验旨在探讨仔猪饲料中常用蛋白原料的蛋白消化速率、肽含量和功能,为准确评价蛋白原料的营养价值提供参考。选取秘鲁蒸汽鱼粉、去皮豆粕、大豆浓缩蛋白、膨化大豆、酵母水解物、发酵豆粕、大豆酶解蛋白7种常用蛋白原料,通过体外胃-胰蛋白酶两步法设定不同消化时间点,采集消化液测定氮含量,计算蛋白消化速率。选取秘鲁蒸汽鱼粉、酵母水解物、发酵豆粕、大豆酶解蛋白4种蛋白原料,通过HPLC色谱法、ABTS法、持钙能力检测法分别对150~1 000 Da肽含量、总抗氧化能力、持钙能力进行分析。结果显示:秘鲁蒸汽鱼粉、大豆浓缩蛋白、发酵豆粕在240 min消化率显著提高(P<0.05);去皮豆粕在150 min消化率显著提高(P<0.05);膨化大豆在210 min消化率显著提高(P<0.05);酵母水解物在120 min消化率显著提高(P<0.05);大豆酶解蛋白在60 min消化率显著提高(P<0.05)。大豆酶解蛋白、酵母水解物在90 min时,消化率显著高于其他5种蛋白原料(P<0.05)。4种饲用蛋白原料150~1 000 Da肽含量由大致小、总抗氧化及持钙能力由强至弱依次为酵母水解物、大豆酶解蛋白、发酵豆粕、鱼粉。研究表明,通过胃-胰蛋白酶两步法、HPLC色谱法、ABTS法、持钙检测法可以准确测定蛋白原料体外蛋白消化速率、小分子肽含量、总抗氧化能力、持钙能力,深加工酶解的蛋白原料消化速率快、功能性强,具有更好的应用价值。 The experiment was to study the protein digestion rate, peptide content and function of common protein raw materials in suckling piglets feed, to accurately evaluate the nutritional value of protein raw materials. Seven common protein raw materials including peruvian steam fish meal, dehulled soybean meal, soybean protein concentrate, expanded soybean, yeast hydrolysate, fermented soybean meal and soybean enzymatic hydrolysate were selected to set different digestion time points by in vitro gastric trypsin two-step method. The nitrogen content of digestive fluid was collected and the protein digestion rate was calculated. Four kinds of protein raw materials, including peruvian steam fish meal, yeast hydrolysate, fermented soybean meal and soybean enzymolysis protein, were selected. The contents of 150~1 000 Da peptide, total antioxidant capacity and calcium holding capacity were analyzed by HPLC, ABTS and calcium holding capacity detection method. The results showed that the digestibility of peruvian steam fish meal, soybean protein concentrate and fermented soybean meal increased significantly at 240 min(P<0.05), the digestibility of dehulled soybean meal was significantly increased at 150 min(P<0.05), the digestibility of puffed soybean was significantly increased at210 min(P<0.05), the digestibility of yeast hydrolyzate was significantly increased at 120 min(P<0.05), and the digestibility of soybean enzymolyzed protein was significantly increased at 60 min(P<0.05). The digestibility of soybean enzymolyzed protein and yeast hydrolyzate at 90 min was significantly higher than that of the other five protein raw materials(P<0.05). The contents of 150~1 000 Da peptides in the four feed protein raw materials from large to small, and the total antioxidant and calcium holding capacity from strong to weak were yeast hydrolyzate, soybean enzymolyzed protein, fermented soybean meal and fish meal.
