摘要
以青海互助地区主栽的7个不同品种草莓匍匐茎茎尖为外植体,利用MS基本培养基添加适宜浓度的6-BA、GA_(3)或IBA,探索草莓茎尖组织培养获得脱毒苗的方法。结果表明:以草莓茎尖组织培养技术培育出的草莓脱毒苗,幼苗移栽后生长、开花和结果正常。草莓茎尖分化培养基MS+0.5mg/L 6-BA+0.2mg/L GA_(3)适合“红颜”等4个品种茎尖生长;增殖培养基MS+0.5mg/L 6-BA+0.2mg/L GA_(3)适合草莓“红颜”、“通州公主”、“小白”、“京泉香”和“章姬”5个品种幼苗增殖,增殖系数均在7以上。生根培养基MS+0.25mg/L IBA短期内各品种生根较快,但生根超过20d时幼苗根部生长异常,还需进一步试验找到适宜培养基。
The stolon tips of 7 different strawberry varieties mainly planted in Huzhu area of Qinghai Province were used as explants,and the MS basic medium was added with appropriate concentration of 6-BA,GA3 or IBA to explore the method of obtaining virus-free seedlings by tissue culture of strawberry stolon tips.The results showed that the virus-free strawberry seedlings cultivated by strawberry stem tip tissue culture technology had normal growth,flowering and fruit after transplanting.Strawberry shoot tip differentiation medium MS+0.5 mg/L6-BA+0.2 mg/L GA_(3) was suitable for shoot tip growth of four varieties such as‘Benihoppe’strawberry(Fragaria×ananassa Duch);The proliferation medium MS+0.5 mg/L6-BA+0.2 mg/LGA_(3) was suitable for the proliferation of strawberry‘Benihoppe’,‘TongZhou Princess’,‘Xiaobai’,‘jingquanxiang’and‘Zoji’,and the proliferation coefficients were more than 7.Rooting medium MS+0.25mg/L IBA could root faster in a short time,but the root growth of seedlings was abnormal when rooting for more than 20 days.Further experiments were needed to find a suitable medium.
