摘要 目的 建立低温冷冻条件下消毒剂杀灭效果的测定方法。方法 选用大肠杆菌和金黄色葡萄球菌两种代表性的菌种,进行菌悬液配方和染菌载体优化,以市场流通的冷链低温消毒剂作为样品,进行低温杀灭效果的测定。结果 在-40℃、-20℃、4℃、20℃温度下,常温消毒剂低温试验结冰无法进行杀灭试验;未优化的菌悬液低温下平衡会结冰影响试验结果,染菌载体菌落回收数明显降低;在配方优化后,低温消毒剂在相应低温下用悬液定量法作用10 min后对大肠杆菌和金黄色葡萄球菌的杀灭对数值均>5.00;载体喷雾定量法作用10 min后对大肠杆菌和金黄色葡萄球菌的杀灭对数值均>3.00。结论 该方法的研究建立适用于新冠疫情下低温冷链低温消毒剂的杀灭试验,为其他实验室的低温模拟杀灭试验提供一定的参考价值。 Objective To establish a method for determining the killing effect of disinfectants under low temperature freezing conditions. Methods Two representative strains of Escherichia coli and Staphylococcus aureus were selected to optimize the formula of bacterial suspension and the carrier of bacterial contamination. The cold-chain low-temperature disinfectant in market circulation was used as the sample to determine the low-temperature killing effect. Results At the temperatures of -40 ℃,-20 ℃, 4 ℃, 20 ℃, the normal temperature disinfectant is frozen in the low temperature test, and the killing test cannot be carried out;the balance of non optimized bacterial suspension will freeze at low temperature, which will affect the test results, and the colony recovery number of infected carrier will be significantly reduced;after optimizing the formula, the logarithm of killing Escherichia coli and Staphylococcus aureus by the suspension quantitative method at the corresponding low temperature for 10 minutes was >5.00;the logarithm value of carrier spray quantitative method for killing Escherichia coli and Staphylococcus aureus after 10 min was >3.00. Conclusion The study of this method is suitable for the killing test of lowtemperature cold chain low-temperature disinfectant under COVID-19, and provides a certain reference value for the low-temperature simulation killing test in other laboratories.
