摘要
目的:探讨黏附分子δ-catenin能否通过促进肿瘤细胞的干性特征以及端粒酶的活性而参与肺癌的侵袭进展。方法:应用免疫组化检测肺癌组织中δ-catenin、癌症干细胞标志物(ABCG2、Nanog)、端粒酶逆转录酶(telomerase reverse transcriptase,TERT)的表达情况。利用流式细胞术在肺癌细胞系中分选肿瘤干细胞,借助Western Blotting、侵袭实验分别检测在肿瘤干细胞中改变δ-catenin蛋白表达后ABCG2、Nanog、TERT和肺癌细胞侵袭能力的变化。结果:δ-catenin、ABCG2、Nanog、TERT蛋白在肺癌组织中表达显著增高,且四种蛋白的阳性表达与患者的不良预后密切相关。在肿瘤干细胞中改变δ-catenin表达水平后,ABCG2、Nanog、TERT蛋白表达与肺癌细胞侵袭能力也相应地改变。结论:δ-catenin可能通过增强肿瘤细胞的干性特征和端粒酶活性而促进肺癌的侵袭进展。
Objective:To investigate whetherδ-catenin can participate in the invasion and progression of lung cancer by promoting the stem cells characteristics of tumor cells and telomerase activity.Methods:Immunohistochemistry was used to detect the expression ofδ-catenin,cancer stem cell marker(ABCG2 Nanog),telomerase reverse transcriptase(TERT)in lung cancer tissues.Furthermore,cancer stem cells were selected in lung cancer cell lines by flow cytometry.The expression of ABCG2,Nanog,TERT and the invasion ability of tumor cells was detected by Western Blotting and cell invasion assay after changingδ-catenin expression in cancer stem cells.Results:The expression ofδ-catenin,ABCG2,Nanog,TERT protein were significantly increased in lung cancer tissues,and positive expression of the four proteins were closely associated with poor prognosis in patients.The expression of ABCG2,Nanog,TERT and invasive ability of tumor cells were changed accompanied with changedδ-catenin expression in cancer stem cells.Conclusion:δ-catenin may promote tumor invasion and progression by enhancing the stem cells characteristics and telomerase activity of lung cancer cells.
