摘要
【目的】探究确山黑猪HOXA5基因的组织表达情况和HOXA5蛋白的结构功能。【方法】利用实时荧光定量PCR(real-time quantitative PCR,qRT-PCR)技术检测HOXA5基因在确山黑猪各组织中的表达情况,并与大白猪皮下脂肪和背最长肌进行对比;以确山黑猪皮下脂肪组织为cDNA模板,克隆确山黑猪HOXA5基因编码(coding sequence,CDS)区序列,利用生物信息学软件分析确山黑猪HOXA5基因CDS区序列及其编码蛋白质的结构和功能。【结果】HOXA5基因在确山黑猪肺脏中表达量最高,其次是肝脏和皮下脂肪,且显著高于其他组织。同时,HOXA5基因在确山黑猪皮下脂肪组织中表达量显著高于大白猪。通过对确山黑猪HOXA5基因进行克隆及生物信息学分析,获得的确山黑猪HOXA5基因CDS区全长813 bp,编码270个氨基酸,与牛、绵羊、马、黑猩猩、小鼠、人和鸡的同源性分别为99.1%、99.0%、98.8%、97.8%、97.4%、95.6%和81.3%。确山黑猪HOXA5蛋白分子质量为29.28 kD,主要位于细胞核内,属于酸性、亲水性蛋白,且不具有跨膜结构和信号肽;在二级结构和三级结构中,HOXA5蛋白由α-螺旋、延伸链、β-转角和无规则卷曲组成,为混合型蛋白,以无规则卷曲为主。【结论】HOXA5基因作为调控猪脂肪沉积的候选基因,在确山黑猪品种的改良中具有重要意义。
【Objective】The objective was to explore the tissue expression of HOXA5 gene and the structure and function of HOXA5 protein in Que’shan black pigs.【Method】Real-time quantitative PCR(qRT-PCR)was used to detect the expression difference of HOXA5 gene in Que’shan black pigs tissues,and the results were compared with subcutaneous fat and longissimus dorsi muscle of Large white pigs;The subcutaneous adipose tissue of Que’shan black pigs was used as a cDNA template to clone the HOXA5 gene coding sequence(CDS)region.Bioinformatics analysis software was used to analyze the sequence of the CDS region of HOXA5 gene in Que’shan black pigs and the structure and function of the encoded protein.【Result】The expression of HOXA5 gene was the highest in the lung of Que’shan black pigs,followed by liver and subcutaneous fat,and was significantly higher than that in other tissues.Meanwhile,the HOXA5 gene expression in subcutaneous adipose tissue of Que’shan black pigs was significantly higher than that in Large white pigs.Through cloning and bioinformatics analysis of HOXA5 gene in Que’shan black pigs,the CDS region of porcine HOXA5 gene in Que’shan black pigs was fould to be 813 bp in length,encoding 270 amino acids.The homology was 99.1%,99.0%,98.8%,97.8%,97.4%,95.6%and 81.3%with bos taurus,ovis aries,equus caballus,pan troglodytes,mus musculus,homo sapiens and gallus gallus,respectively.The molecular weight of HOXA5 protein from Que’shan black pigs was 29.28 kD,which was mainly located in the nucleus and was acidic and hydrophilic,without transmembrane structure and signal peptide.In the secondary and tertiary structure,HOXA5 protein is composed ofα-helix,extended chain,β-turn,and random coils.HOXA5 protein is a mixed type protein,mainly with random coils.【Conclusion】As a candidate gene to adjust and control the fat deposition of pigs,HOXA5 gene,has very important meaning in improvement of Que’shan black pigs.
