摘要
目的:采用酸水解系统-索氏提取仪(仪器法)测定食品中的总脂肪含量,并与国家标准方法对比。方法:样品包裹在滤袋中,并置于酸水解系统,加入盐酸按照程序设定进行酸水解,烘干后进行索氏提取,干燥称重获得结果。结果:经优化实验条件后,仪器法测定食品总脂肪的重复性相对标准偏差(n=6)为0.57%~3.86%,经配对样本t检验,仪器法与国家标准方法测定结果无显著性差异t=1.221,P=0.253(P>0.05),测定结果基本一致。结论:应用酸水解系统-索氏提取仪检测食品中总脂肪的自动化程度高,操作简便,工作效率高,方法的准确度和精确度高,安全性高,值得推广应用。
Objective:To determine total fat content in food by hydrochloric acid hydrolysis system-soxhlet extractor(Instrumental method),and compare with national standard method.Method:The sample eas wrapped in the filter bag and placed in the hydrochloric acid hydrolysis system.Hydrochloric acid was added and started to acid hydrolysis according to the program.After drying,soxhlet extraction was carried out,dried and weighed to obtain the results.Result:The relative standard deviation of repeatability were 0.57%~3.86%(n=6)when determining total fat in food by instrumental method.By paired sample t-test,there was no significant difference between instrument method and national standard method t=1.221,P=0.253(P>0.05),the determination resultswere basically consistent.Conclusion:The application of hydrochloric acid hydrolysis system-soxhlet extractor to detect total fat in food has the advantages of high automation,simple operation,high efficiency,high accuracy and accuracy and high safety,which was worthy promoting and applying.
