摘要 目的:研究鹿茸多糖对MC3T3-E1成骨细胞与BMSCs骨髓间充质干细胞增殖与分化的影响。方法:用最佳起效浓度的鹿茸多糖提取液培养两种细胞24 h、48 h、72 h后,用CCK-8检测细胞的增殖活性,采用ELISA法检测细胞内ALP蛋白酶活性。结果:与空白对照相比,以鹿茸多糖提取液培养两种细胞24 h、48 h、72 h后,细胞活性均明显提高;培养MC3T3-E1成骨细胞24 h后,细胞内ALP蛋白酶活性增高,具有显著性差异。结论:鹿茸多糖提取液能够通过促进成骨细胞与骨髓间充质干细胞的增殖,以及上调成骨细胞内ALP蛋白酶活性等环节起到抗骨质疏松作用。 Objective To study the effect of antler polysaccharide on the proliferation and differentiation of MC3T3-E1 osteoblasts and BMSCs bone marrow mesenchymal stem cells.Method After cultivating two kinds of cells with the optimal concentration of velvet antler polysaccharide extract for 24 h,48 h,72 h,Use CCK-8 to detect cell proliferation activity,ELISA method was used to detect the ALP protease activity in the cells.Results Compared with the blank control group,after culturing the two kinds of cells with velvet antler polysaccharide extract for 24 h,48 h,72 h,the cell viability was significantly improved,and there was a very significant difference.After culturing MC3T3-E1 osteoblasts for 24 hours,the ALP protease activity in the cells increased,with significant differences.Conclution Antler polysaccharide extract can play an anti-osteoporosis effect by promoting the proliferation of osteoblasts and bone marrow mesenchymal stem cells,and up-regulating the ALP protease activity in osteoblasts.
