摘要
目的分析淫羊藿苷影响抑郁动物模型神经内分泌系统功能及其相关机制。方法选择清洁级SD小鼠80只,分为对照组、模型组、低剂量组及高剂量组,其中模型组、低剂量组及高剂量组建立社交应激抑郁模型,对照组不予处理。观察并比较各组小鼠社区区域停留时间、体质量及水平穿越格数,于心脏采血,采用酶联免疫吸附试验检测血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。取下丘脑,按照促肾上腺皮质激素释放激素(CRF)放免试剂盒说明书检测各组小鼠CRF水平,取垂体及肾上腺,按照促肾上腺皮质激素(ACTH)、肾上腺皮质肌动蛋白(CORT)试剂盒说明书检测各组小鼠ACTH、CORT水平。放射性配基检测糖皮质激素受体(GR)最大结合容量,Western blot检测GR蛋白表达。结果模型组、低剂量组、高剂量组小鼠水平穿越格数、社区区域停留时间及体质量明显低于对照组,低剂量组和高剂量组小鼠水平穿越格数、社区区域停留时间及体质量明显高于模型组,且上述指标随剂量的增加而升高(P<0.05)。模型组、低剂量组、高剂量组小鼠IL-6、TNF-α水平明显高于对照组,低剂量组和高剂量组小鼠IL-6、TNF-α水平明显低于模型组,且随剂量的增加而降低(P<0.05)。模型组、低剂量组、高剂量组小鼠CRF、ACTH、CORT水平明显高于对照组,低剂量组和高剂量组小鼠CRF、ACTH、CORT水平明显低于模型组,且随剂量的增加而降低(P<0.05)。模型组、低剂量组、高剂量组小鼠GR结合力明显低于对照组,低剂量组和高剂量组小鼠GR结合力明显高于模型组,且随剂量的增加而升高(P<0.05)。模型组、低剂量组、高剂量组小鼠GR蛋白水平明显低于对照组,低剂量组和高剂量组小鼠GR蛋白水平明显高于模型组,且随剂量的增加而升高(P<0.05)。结论淫羊藿苷可良性调整抑郁小鼠社交行为及神经内分泌功能,降低小鼠慢性炎性反
Objective To analyze the effect of icariin on neuroendocrine system in the depression animal model and its related mechanism.Methods Eighty clean grade SD mice were divided into the control group,model group,low-dose group and high-dose group.Among them,the model group,low-dose group and high-dose group were established the social contact stress-induced depression model,and the control group was not treated.The residence time in community area,body weight and number of horizontal crossing grids of mice were observed and compared among the groups.Blood samples were collected from the heart.Serum interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)levels were detected by enzyme-linked immunosorbent assay(ELISA).The level of corticotropin releasing hormone(CRH)in hypothalamus was detected according to the instructions of CRF radioimmunoassay kit.The levels of ACTH and CORT in pituitary and adrenal gland were detected according to the instructions of ACTH and CORT kit.The maximum binding capacity of glucocorticoid receptor was detected by radioligand assay,and the expression of glucocorticoid receptor protein was detected by Western blot.Results The number of horizontal crossing grids,residence time in community area and body weight in the model group,low-dose group and high-dose group were significantly lower than those in the control group,while the number of horizontal crossing grids,residence time in community area and body weight in the low-dose group and high-dose group were significantly higher than those in the model group,moreover the above indicators were increased with the dose increase(P<0.05).The levels of IL-6 and TNF-αin the model group,low dose group and high dose group were significantly higher than those in the control group,while the levels of IL-6 and TNF-αin the low-dose group and high-dose group were significantly lower than those in the model group,moreover which were decreased with the dose increase(P<0.05).The levels of CRF,ACTH and CORT in the model group,low-dose group and high-dose gr
