雷公藤多苷抗大鼠实验性牙周炎的研究 被引量:5

Effect of tripterygium wilfordii polyglycosides on experimental periodontitis in rats

摘要 目的 观察雷公藤多苷对大鼠实验性牙周炎的抑制情况,并探讨其可能的机制,为临床治疗牙周疾病提供新思路。方法 选取六周龄雄性SD大鼠随机分为4组;正常对照组(NL)、单纯结扎组(LC)、结扎+盐水组(L+NaCl)、结扎+雷公藤多苷组(L+TWP),结扎同时给药处理,7天后处死,取样,对其牙周临床指标、炎症细胞因子mRNA水平、血清中肌酐(creatinine, Cr)、丙氨酸氨基转移酶(alanine aminotransferase, ALT)、天冬氨酸氨基转移酶(aspartate aminotransferase, AST)、肝脏及牙龈组织学结构进行检测。结果 各组间简化软垢指数(debris index simplified, DIS)无统计学差异(P>0.05),但L+TWP组牙龈出血指数(sulcus bleeding index, SBI)、牙周袋深度(probing depth, PD)及临床附着丧失水平(clinical attachment level, CAL)值较L+NaCl显著降低(P<0.05);qRT-PCR测得L+TWP组白细胞介素1β(interleukin1β, 1L-1β),白细胞介素6(interleukin 6,1L-6)和肿瘤坏死因子-a(tumor necrosis factor-α, TNF-α)mRNA的表达显著低于L+NaCl组(P<0.05);各组大鼠血清中ALT、AST和Cr水平无统计学差异(P>0.05);组织学染色示L+TWP组炎症细胞的表达接近正常组;牙周组织亚甲蓝染色示LC组及L+NaCl组牙槽骨吸收较L+TWP组显著。结论 雷公藤多苷可以通过调控多种炎症细胞因子的表达抑制牙龈组织炎症和骨吸收水平,有可能成为抑制牙周炎的有效辅助药物。 Objective Observe the inhibitory effect of tripterygium wilfordii polyglycosides on experimental periodontitis in rats and explore its possible mechanism to provide new ideas for clinical treatment of periodontal diseases.Methods Six-week-old male SD rats were randomly divided into 4 groups;normal control group(NL), ligation control group(LC), ligation control+NaCl group(L+NaCl), ligation control + tripterygium wilfordii polyglycosides group(L+TWP), ligated and administered at the same time. Rats were sacrificed after 7 days. Samples were taken to test the periodontal clinical indicators, inflammatory cytokine mRNA levels, ALT, AST, Cr, liver and gingival histological structure. Results The value of DIS was not statistically different in all groups(P>0.05), but the SBI, PD and CAL value of the L+TWP group was significantly lower than that of L+NaCl(P<0.05);the qRTPCR experiment showed that the expression of IL-1β, IL-6 and TNF-α mRNA in the L +TWP group was significantly lower than that in L+NaCl group(P<0.05);There was no statistical difference in the levels of ALT,AST and Cr in the serum of rats in each group(P >0.05);HE staining showed that the expression of inflammatory cells in the L +TWP group was close to the normal group. Conclusion Tripterygium wilfordii polyglycosides can inhibit the level of inflammation and bone resorption in gum tissues by regulating the expression of a variety of inflammatory cytokines, and may become an effective adjuvant drug for inhibiting periodontitis.
