摘要
目的:探讨重组人AMH对人卵巢癌细胞株SKOV3迁移和侵袭的影响。方法:将浓度为200nmol/L重组人AMH加入培养的卵巢癌细胞SKOV3中,培养48h,作为观察组。不经过任何处理的卵巢癌细胞SKOV3作为对照组。观察组和对照组都平行设置3个复孔。采用细胞创伤愈合实验检测两组细胞迁移能力的变化,采用Transwell小室体外侵袭实验检测两组细胞侵袭能力的变化。结果:观察组细胞的迁移速度是(2.99±0.04)μm/h,观察组细胞穿过膜的细胞数为(15.00±1.00)个,阴性对照组细胞的迁移速度为(9.02±0.06)μm/h,阴性对照组穿过膜的细胞数为(39.70±1.50)个,观察组细胞迁移速度明显慢于对照组(P<0.05)。观察组细胞穿过膜的细胞数目明显少于阴性对照组,两组相对侵袭指数差异有显著性(P<0.05)。结论:重组人AMH可抑制人卵巢癌细胞株SKOV3的迁移和侵袭能力,为卵巢癌治疗提供一种新的思路。
Objective:To investigate the effect of recombinant human AMH on the migration and invasion of human ovarian cancer cell line SKOV3.Methods:Recombinant human AMH with a concentration of 200nmol/L was added to the cultured ovarian cancer cell SKOV3 and cultured for 48 hours as the observation group.Ovarian cancer cell SKOV3 without any treatment was used as the control group.Three compound holes were set in parallel in the observation group and the control group.The changes of cell migration ability of the two groups were detected by cell wound healing experiment,and the changes of cell invasion ability of the two groups were detected by Transwell chamber invasion experiment in vitro.Results:The migration rate of cells in the observation group was(2.99±0.04)μm/h,the number of cells passing through the membrane in the observation group was 15.00±1.00,and the migration rate of cells in the negative control group was(9.02±0.06)μm/h,the number of cells passing through the membrane in the negative control group was 39.70±1.50,and the migration speed of cells in the observation group was significantly slower than that in the control group(P<0.05).The number of cells passing through the membrane in the observation group was significantly less than that in the negative control group,the difference in the relative invasion index was significant between the two groups.Conclusion:Recombinant human AMH can inhibit the migration and invasion of human ovarian cancer cell line SKOV3,which provides a new idea for the treatment of ovarian cancer.
