赤芍总苷通过调控miR-200b-3p/FSCN1轴抑制卵巢癌细胞增殖、侵袭和迁移 被引量:2

Total paeony glycosides inhibit the proliferation, invasion and migration of ovarian cancer cells by regulating the miR-200b-3p/FSCN1 axis

摘要 目的探讨赤芍总苷对卵巢癌增殖、侵袭和迁移的影响及可能机制。方法体外培养卵巢癌细胞SKOV3,经不同剂量(50μg/mL、100μg/mL和200μg/mL)赤芍总苷干预24 h、或转染miR-200b-3p模拟物至SKOV3细胞、或200μg/mL的赤芍总苷干预转染miR-200b-3p抑制剂的SKOV3细胞24 h后,细胞计数试剂盒-8(CCK-8)法和克隆形成实验检测细胞增殖,划痕实验检测细胞迁移,Transwell检测细胞侵袭,实时定量聚合酶链反应(RT-qPCR)法检测miR-200b-3p表达,Western blot检测细胞中肌动蛋白成束蛋白(FSCN1)表达。双荧光素酶报告基因实验验证FSCN1和miR-200b-3p调控关系。结果赤芍总苷可降低SKOV3细胞光密度(OD)值、克隆形成数、划痕愈合率、侵袭数及细胞中FSCN1蛋白表达(P<0.05),而促进miR-200b-3p表达(P<0.05),且呈剂量依赖性。过表达miR-200b-3p可降低SKOV3细胞OD值、克隆形成数、划痕愈合率、侵袭数及细胞中FSCN1蛋白表达(P<0.05)。miR-200b-3p靶向负调控FSCN1。敲减miR-200b-3p减弱赤芍总苷对SKOV3细胞增殖、迁移和侵袭的抑制作用(P<0.05)。结论赤芍总苷可能通过调控miR-200b-3p/FSCN1轴抑制卵巢癌细胞增殖、迁移和侵袭。 Objective To investigate the effect of total paeony glucosides on the proliferation, invasion and migration of ovarian cancer and its possible mechanism. Methods Ovarian cancer cells SKOV3 were cultured in vitro. After SKOV3 cells were treated with different doses of total paeony glycosides(50 μg/mL, 100 μg/mL and 200 μg/mL) for 24 h, or miR-200 b-3 p mimics were transfected into SKOV3 cells, or SKOV3 cells transfected with miR-200 b-3 p inhibitor were treated with 200 μg/mL total paeony glucosides for 24 h, cell counting kit-8(CCK-8) method and clone formation test were used to detect cell proliferation, scratch test was used to detect cell migration, Transwell was used to detect cell invasion, real-time quantitative polymerase chain reaction(RT-qPCR) was used to detect the expression of miR-200 b-3 p, and Western blot was used to detect the protein expression of fascin actin-bundling protein 1(FSCN1). The dual luciferase reporter gene experiment verified the regulatory relationship between FSCN1 and miR-200 b-3 p. Results The total glucosides of paeony reduced the optical density(OD) value of SKOV3 cells, the number of clone formation, the wound healing rate, the number of invasion and the protein expression of FSCN1(P<0.05), while promoted the expression of miR-200 b-3 p(P<0.05), and it were dose dependent. Overexpression of miR-200 b-3 p reduced the OD value of SKOV3 cells, the number of clone formation, the wound healing rate, the number of invasion and the protein expression of FSCN1(P<0.05). miR-200 b-3 p negatively regulated the protein expression of FSCN1. Knockdown of miR-200 b-3 p attenuated the inhibitory effect of total paeony glycosides on the proliferation, migration and invasion of SKOV3 cells(P<0.05). Conclusion Total paeony glycosides may inhibit the proliferation, migration and invasion of ovarian cancer cells by regulating the miR-200 b-3 p/FSCN1 axis.
