摘要
为了解我国山东省、河南省、河北省驴群中马动脉炎病毒(EAV)和马疱疹病毒8型(EHV-8)的感染现状,于2020年从3个省份的屠宰场和驴场收集驴血样品共计860份。通过RT-PCR和PCR方法分别对血样进行EAV和EHV-8检测,统计其阳性率和混合感染率,并分析其流行情况;同时,对3个省份EAV和EHV-8阳性样品的ORF7基因和ORF70部分基因序列进行克隆、测序及遗传进化分析。结果显示,EAV的阳性率为5.7%(49/860),EHV-8的阳性19.7%(169/860),混合感染率为1.9%(16/860)。分析显示,3个省份扩增的EAV ORF7基因测序结果均与经典毒株EAV Bucyrus同源性最高,亲缘关系最近;河北EHV-8 ORF70基因与EHV-8 IR/2010/16亲缘关系最近,河南EHV-8 ORF70基因与EHV-8 IR/2010/47亲缘关系最近,山东EHV-8 ORF70基因与EHV-8 wh亲缘关系最近。论文首次报道EAV和EHV-8在驴群感染情况,为下一步防控EAV和EHV-8奠定了基础。
In order to understand the infection status of EAV and EHV-8,860 blood samples were collected from slaughterhouses and donkey farms of different areas in Shandong,Hebei and Henan provinces and detected for EAV and EHV-8 by RT-PCR/PCR with specific primers in 2020.The positive rate and co-infection rate were calculated respectively.The EAV ORF7 gene and EHV-8 ORF70 genes from positive blood samples were cloned into pTOPO vector,sequenced and analyzed for genetic evolution.The results showed that positive rate of EAV was 5.7%(49/860),positive rate of EHV-8 was 19.7%(169/860),and co-infective positive rate was 1.9%(16/860).Bioinformatics analysis showed that the ORF7 gene sequences of EAV from 3 provinces share highest homology and the closest genetic relationship with the Bucyrus strain.The ORF70 gene sequence of EHV-8 from Hebei province was most closely related to EHV-8 IR/2010/16,the ORF70 gene sequence of EHV-8from Henan province was most closely related to EHV-8 IR/2010/47,and the ORF70 gene sequence of EHV-8 from Shandong province was most closely related to EHV-8 wh.Overall,our study first reported the prevalences of EAV and EHV-8 in donkeys of Shandong,Hebei and Henan provinces to lay a foundation for further prevention and control of EAV and EHV-8.
