顺铂诱导急性肾损伤差异基因的分析和鉴定 被引量:1

摘要 目的:分析与鉴定顺铂诱导急性肾损伤(Cisplatin induced acute kidney injury,Cis-AKI)的关键差异基因。方法:选取公共基因表达数据库(Gene Expression Omnibus,GEO)中Cis-AKI大鼠的微阵列测序数据集进行分析,通过R软件中的limma包分析Cis-AKI的差异基因(Differentially expressed genes,DEGs),对差异基因进行基因的本体论(Gene Ontology,GO)分析、京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析。使用MCODE模块和CytoHubba模块筛选蛋白质互作网络(Protein-Protein Interaction Networks,PPI)中的关键差异基因。结果:与对照组相比,Cis-AKI组共有119个DEGs,其中上调基因62个,下调基因57个。DEGs主要富集在细胞周期和昼夜节律等通路上。ARNTL、CLOCK、CRY1、DBP、PER3、NR1D1、CCNG1、FOS、BTG2和AEN基因被鉴定为是Cis-AKI中的关键差异基因。这些关键差异基因主要富集在p53信号通路、细胞周期和昼夜节律等相关通路。结论:利用生物信息学方法对Cis-AKI差异表达基因的分析,鉴定出ARNTL、CLOCK、CRY1、DBP、PER3、NR1D1、CCNG1、FOS、BTG2和AEN等基因是Cis-AKI中的关键差异基因。 Objective:To analyze and identify the key differential genes of cisplatin induced acute kidney injury(Cis-AKI). Methods:The microarray sequencing dataset of cisplatin induced acute kidney injury(Cis-AKI)in the public gene expression database(Gene Expression Omnibus,GEO)was selected for analysis,and analyzed by the limma package in R software. Differentially expressed genes(DEGs)of Cis-AKI,Gene Ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis were performed on the differential genes. The key differential genes in Protein-Protein Interaction Networks(PPI) were screened using MCODE module and CytoHubba module. Results:Compared with the control group,a total of 119 DEGs in theCis-AKI group included62 upregulated genes and 57 down-regulated genes. DEGs are mainly enriched in the cell cycleand circadian rhythm pathways.ARNTL,CLOCK,CRY1,DBP,PER3,NR1D1,CCNG1,FOS,BTG2 and AEN genes were identified as key differential genes in Cis-AKI. These key differential genes were mainly enriched in p53 signaling pathway,cell cycle and circadian rhythm pathways. Conclusion:Through the analysis of differentially expressed genes in Cis-AKI using bioinformatics methods,genes such as ARNTL,CLOCK,CRY1,DBP,PER3,NR1D1,CCNG1,FOS,BTG2 and AEN were identified as key differential genes in Cis-AKI.
