初乳添加剂对新生羔羊血清指标和免疫球蛋白G吸收效率的影响

Effects of Colostrum Additives on Serum Indexes and Immunoglobulin G Absorption Efficiency of Newborn Lambs

摘要 本试验旨在研究不同初乳添加剂对新生羔羊血清指标和免疫球蛋白G(IgG)吸收效率的影响。选取60只初生重[(3.52±0.31)kg]相近、健康状况良好的新生湖羊公羔,随机分为6组,每组10只,分别为对照组、抗生素组、甘露寡糖组、甲酸组、乙二胺四乙酸(EDTA)组和西沙必利组,各组羔羊分别将生理盐水、抗生素、甘露寡糖、甲酸、EDTA和西沙必利加入到100 mL牛初乳中混合均匀后于羔羊出生后1 h通过口腔插管法饲喂羔羊,羔羊随母哺乳。于羔羊出生后24 h时颈静脉采血,离心收集血清,血清样品分别测定生化指标、抗氧化指标、炎症指标、肠道通透性指标和免疫指标,采用单向免疫扩散法检测牛源IgG和羊源IgG的含量,计算IgG吸收效率。结果显示:抗生素组和甘露寡糖组羔羊血清葡萄糖(GLU)含量显著高于对照组(P<0.05);各试验组羔羊血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和总抗氧化能力(T-AOC)均显著低于对照组(P<0.05),血清丙二醛(MDA)含量均显著高于对照组(P<0.05);各试验组羔羊血清γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)含量均显著高于对照组(P<0.05);各试验组羔羊血清内毒素(LPS)含量和二胺氧化酶(DAO)活性均显著高于对照组(P<0.05);甲酸组、西沙必利组和甘露寡糖组羔羊IgG吸收效率均显著低于对照组(P<0.05),且甲酸组羔羊IgG吸收效率为0;相关性分析表明,新生羔羊IgG吸收效率与血清SOD活性、T-AOC和IgA含量呈显著正相关(P<0.05),与血清MDA、IL-1β和IL-6含量呈显著负相关(P<0.05)。由此可见,初乳中添加抗生素、甘露寡糖、甲酸、EDTA或西沙必利均无法提高新生羔羊的IgG吸收效率;同时,本研究结果提示提高新生羔羊血清抗氧化能力、降低炎症细胞因子含量可能是提高其IgG吸收效率的有效措施。 This experiment was conducted to investigate the effects of different colostrum additives on serum indexes and immunoglobulin G(IgG)absorption efficiency of newborn lambs.Sixty newborn health Hu lambs with the similar birth weight at(3.52±0.31)kg were selected and randomly divided into 6 groups with 10lambs in each group,they were control group,antibiotic group,mannose oligosaccharide group,formic acid group,ethylenediamine tetraacetic acid(EDTA)group and cisapride group,respectively.Normal saline,antibiotics,mannose oligosaccharide,formic acid,EDTA and cisapride mixed with 100 mL of bovine colostrum were fed by oral intubation within 1 h of birth in above groups,respectively.Lambs were lactated with their mothers.Blood samples were collected from the jugular vein 24 h after birth.After centrifugation,serum samples were used to detect biochemical indexes,antioxidant indexes,inflammatory indexes,intestinal permeability indexes and immune indexes.Serum IgG contents from cattle and sheep were measured by single radial immunodiffusion to calculate the absorption efficiency of IgG.The results showed that serum glucose(GLU)content in antibiotic group and mannose oligosaccharide group was significantly higher than that in control group(P<0.05).Serum superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)activities and total antioxidant capacity(T-AOC)in each experimental group were significantly lower than those in control group(P<0.05),and serum malondialdehyde(MDA)content was significantly higher than that in control group(P<0.05).Serum contents of interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)in each experimental group were significantly higher than those in control group(P<0.05).Serum lipopolysaccharide(LPS)content and diamine oxidase(DAO)activity in each experimental group were significantly higher than those in control group(P<0.05).IgG absorption efficiency in cisapride group and mannose oligosaccharide group was significantly lower than that in control gro
