摘要
乙醇的存在会严重干扰HPLC法对竹子乙醇精炼液中甲酸、乙酸和丙酸等小分子脂肪酸和各种降解糖的检测。研究了国产氨基色谱柱检测单糖、C-18色谱柱检测小分子脂肪酸的效果。结果表明,氨基柱在流动相乙腈与水体积比75∶25、进样体积1μL、体积流量1.01 mL/min、时间15 min和温度30℃条件下,葡萄糖、木糖、阿拉伯糖和半乳糖有较好的分离效果;C-18柱在流动相水与乙腈体积比50∶50、体积流量0.6 mL/min、时间15 min和温度25℃条件下,甲酸、乙酸和丙酸能够完全分离。
The presence of ethanol could seriously affect determination of small fatty acids,formic acid,acetic acid,propionic acid and various degraded sugars in ethanol refining liquid of bamboo by HPLC.The contents of monosaccharides and small fatty acids were analyzed by domestic amino and C-18 columns,respectively.The results showed that the amino column owned a excellent seperation performance to glucose,xylose,arabinose and galactose under the conditions of 75∶25 volume ratio of acetonitrile to water,1μL injection volume,1.01 mL/min volume flow at 30℃for 15 min.The C-18 column could completely separate formic acid,acetic acid and propionic acid from liquors under the condition of 50∶50 volume ratio of water to acetonitrile,0.6 mL/min volume flow rate at 25℃for 15 min.
