摘要
【目的】筛选并鉴定对芋头干腐病菌具有显著拮抗作用的芽孢杆菌,为有效防控芋头干腐病提供生防资源。【方法】采用稀释平板分离法从茶树根际土壤中分离芽孢杆菌;以芋头干腐病的优势病原菌茄镰刀菌(Fusarium solani)为靶标菌,采用平板对峙法筛选拮抗菌株,并测定拮抗菌株的抑菌范围;通过形态观察、生理生化特性测定和16S rDNA、gyrA、rpoB基因序列分析,对拮抗菌株进行种类鉴定。【结果】筛选到1株对芋头干腐病菌具有显著抑菌效果的菌株,命名为D-1。该菌株对供试的其他9种植物病原真菌和5种植物病原细菌也有显著的抑菌作用。D-1菌落圆形、乳白色、不透明、表面干燥皱褶、边缘不整齐、中央凹陷;生理生化特性测定结果与对照菌株贝莱斯芽孢杆菌(Bacillus velezensis)FJ17-4的测定结果一致;测定的16S rDNA、gyrA和rpoB 3个基因序列与GenBank中B.velezensis对应序列的同源性均为100%;在分别基于16S rDNA、gyrA和rpoB基因序列构建的系统发育树上,该菌株与贝莱斯芽孢杆菌(Bacillus velezensis)各菌株聚类成一个分支。【结论】根据形态特征、生理生化特性和多基因序列分析结果,鉴定菌株D-1为贝莱斯芽孢杆菌(B.velezensis),该菌株抑菌活性强,抑菌谱广,在植物病害生物防治中具有应用潜力。
[Objective]The objective of this study is to screen and identify a strain in Bacillus with significant antagonistic activity on the pathogen of taro dry rot,thus providing biocontrol resources for effective control of the disease.[Method]Bacillussp.was isolated from tea rhizosphere soil by dilution-plate method.The antagonistic strains were screened by dual culture with Fusarium solani,the dominant pathogen of taro dry rot,as the target fungus,and the antibacterial spectrum of the obtained antagonistic strain was determined.Species of the antagonistic strain was identified by morphological observation,physiological and biochemical characteristics test and 16S rDNA,gyrA and rpoB gene sequence analysis.[Result]A strain with significant antagonistic effect on F.solani was screened and named D-1.It also had significant antimicrobial effect on all other 9 plant pathogenic fungi and 5 plant pathogenic bacteria.Colonies of strain D-1 on NA were round,milky white,opaque,with dry and wrinkled surface,irregular edge,and sunken center.The results of physiological and biochemical characteristics were consistent with those of the reference strain Bacillus velezensis FJ17-4.The sequences of 16S rDNA,gyrA and rpoB of the strain shared the highest homology of 100%with the counterpart of B.velezensis in GenBank.The strain was clustered into a branch with other strains of B.velezensis in the phylogenetic tree of 16S rDNA gene,phylogenetic tree of gyrA gene or phylogenetic tree of rpoB gene.[Conclusion]Based on the morphological features,physiological and biochemical characteristics and 16S rDNA,gyrA and rpoB gene sequence analysis,strain D-1 was identified as B.velezensis.It had strong antagonistic activity,broad antibacterial spectrum,and application potential in the biological control of plant diseases.
