摘要
本试验主要对辛酸法、水稀释法、泊洛沙姆法3种不同提取抗体方法进行了比较,通过观察抗体的物理性状、检测提取液蛋白含量、使用血凝试验与血凝抑制试验检测抗体效价来选择卵黄抗体的最佳方法。结果表明:3种不同提取方法的蛋白含量以泊洛沙姆法最高、辛酸法最低;新城疫和禽流感效价以水稀释法最高、辛酸法最低;抗体溶液体积以水稀释法最高、泊洛沙姆法最低。综合比较试验结果,最终确定水稀释法效果最优。采用该方法提取的卵黄抗体效价最高、易操作,可实现车间大规模生产。
The experiment mainly compared three different extraction methods of antibodies:caprylic acid method,water dilution method and poloxamer method.By observing the physical properties of the antibody,detecting the protein content of the extract,and using the hemagglutination test and the hemagglutination inhibition test to detect the antibody titer,select the best process of the yolk antibody.The results showed that among the three different extraction methods,the protein content was the highest in poloxamer method and the lowest in caprylic acid method;the titers of newcastle disease and avian influenza were the highest in water dilution method and the lowest in caprylic acid method;the volume of antibody solution was the highest in water dilution method and the lowest in poloxamer method.The experimental results were comprehensively compared,and it was finally determined that water dilution method had the best effect.When using this method to extract yolk antibody,the titer was the highest,and it was easy to operate,it could achieve large-scale production workshop.
