高效液相色谱法同时测定饮料中7种防腐剂和2种甜味剂 被引量:7

摘要 目的:建立同时测定饮料中7种防腐剂2种甜味剂的高效液相色谱(High Performance Liquid Chromatography,HPLC)分析方法。方法:选用有代表性的果汁类饮料、碳酸饮料、含乳饮料共3种基质,用甲醇水溶液(70%)提取后,定容过滤,蛋白含量较高的样品中加入蛋白沉淀剂净化后定容过滤待上机。选用Shimadzu VP-ODS(150 mm×4.6 mm,4.6μm)色谱柱,用20 mmol/L乙酸铵和甲醇为混合流动相;检测波长为245 nm;进样量为20μL;流速为1.5 mL/min,用优化后的色谱梯度条件进行洗脱。结果:7种防腐剂和2种甜味剂在0.5~50μg/mL浓度范围内线性关系良好(R>0.9999),方法检出限为0.3~2.0 mg/kg,样品加标实验回收率在85.3%~98.5%,相对标准偏差为2.2%~4.5%。结论:优化后的方法简便、快速、准确和可靠,达到方法学和食品安全国家标准限量的要求,适用于饮料样品中防腐剂和甜味剂的同时检测。 Objective:A High performance liquid chromatography(HPLC)method was developed for rapid determination of 7 preservatives and 2 sweeteners in beverage.Method:Fruit juice,carbonated drink and milk beverage were chosen as experiment materials.The samples were extracted by 70%methanol solution with protein precipitator.Then the liquid was analyzed by HPLC.The analytes were separated on Shimadzu VP-ODS(150 mm×4.6 mm,4.6μm)column using methanol and 20 mmol/L ammonium acetate as mobile phase for gradient elution at 245 nm wave length.And the injection volume was 20μL.Result:The results showed that the 7 preservatives and 2 sweeteners had good linearity(R>0.9999)between 0.5~50μg/mL,the limits of detection range from 0.3 mg/kg to 2.0 mg/kg.The recoveries range from 85.3%to 98.5%.The relative standard deviations range from 2.2%to 4.5%.Conclusion:The optimized method is simple,rapid,accurate and reliable,which can meet the requirements of methodology and national food safety standards.It is suitable for the simultaneous determination of preservatives and sweeteners in beverage samples.
