环泊酚通过Wnt/β-catenin信号通路对帕金森病大鼠的神经保护作用 被引量:15

Neuroprotective effect cyclophenol on Parkinson’s disease rats through Wnt/β-catenin signaling pathway

摘要 目的 探讨环泊酚通过Wnt/β-catenin信号通路对帕金森病模型大鼠的神经保护作用。方法 采用随机抓取的方法将30只大鼠分为对照组、模型组和环泊酚组,每组各10只。模型组和环泊酚组复制帕金森病模型,对照组注射等量葵花油。环泊酚组大鼠ig环泊酚溶液2 mg/kg,对照组和模型组大鼠ig等量生理盐水,1次/d,连续14 d。电镜下观察中脑神经细胞的超微结构,免疫组化染色观察黑质酪氨酸羟化酶(TH)阳性表达,Western blotting法检测Wnt和β-catenin蛋白表达。结果 与模型组比较,环泊酚组神经细胞和线粒体数量增多,TH阳性表达率升高,Wnt5a、磷酸化β-catenin和核内β-catenin蛋白表达量下降,差异均有统计学意义(P<0.05)。结论 中脑神经细胞减少、线粒体功能障碍、TH阳性表达下降以及Wnt/β-catenin信号通路异常激活可能与帕金森病的发生有关,而环泊酚可通过改善神经细胞和线粒体功能、增加TH表达以及抑制Wnt/β-catenin信号通路激活对帕金森病发挥神经保护作用。 Objective To investigate the neuroprotective effect of cyclophenol for Parkinson’s disease rats through Wnt/β-catenin signaling pathway. Methods 30 Rats were divided into control group, model group, and cyclophenol group according to random grab method,10 rats in each group. The Parkinson’s disease model was established in model group and cyclophenol group, and the control group was injected with the same amount of sunflower oil. Then the cyclophenol group was ig administered with cyclophenol solution(2 mg/kg), and the control group and model group were ig administered with the same amount of normal saline, once daily for 14 d. The ultrastructure of midbrain neurons was observed under electron microscopy, positive expression of tyrosine hydroxylase(TH) in substantia nigra was stained by immunohistochemistry, and Wnt and β-catenin proteins expression were detected by Western blotting method. Results Compared with the model group, the number of nerve cells and mitochondria in the cyclophenol group were increased. The positive expression rate of TH in cyclophenol group was higher than that in model group, and Wnt5 a,phosphorylate β-catenin, and intranuclear β-catenin proteins were decreased, with statistically significant differences(P < 0.05).Conclusion Neurocytopenia, mitochondrial dysfunction, less TH positive expression, and abnormal activation of Wnt/β-catenin signaling pathway in midbrain may be related to the occurrence of Parkinson’s disease, and cyclophenol can improve the function of nerve cells and mitochondria, increase TH expression and inhibit activation of Wnt/β-catenin signaling pathway, which plays a neuroprotective role in Parkinson’s disease.
