摘要
目的研究贯叶金丝桃提取物(HPE)对高原肺水肿(HAPE)大鼠的保护作用。方法80只Wistar大鼠随机分为常氧空白组(NG组),缺氧模型组(HG组),贯叶金丝桃提取物低(HPE-L,100 mg/kg)、中(HPE-M,200 mg/kg)、高(HPE-H,400 mg/kg)剂量组,金丝桃苷(HYP)低(HPE-L,50 mg/kg)、高(HYP-H,100 mg/kg)剂量组和地塞米松(DXM,4 mg/kg)组,每组10只。连续灌胃给药7 d,第4天起除NG组外,其余各组均置于模拟海拔7500 m的高原环境72 h,建立大鼠HAPE模型。观察大鼠肺组织病理变化;测定肺含水量,丙二醛(MDA)、过氧化氢(H_(2)O_(2))、还原型谷胱甘肽(GSH)含量和总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)活力及炎性因子[白细胞介素-1β(IL-1β)、IL-6、血管内皮生长因子(VEGF)和肿瘤坏死因子-α(TNF-α)]的含量。结果与NG组比较,HG组大鼠肺组织充血水肿,肺泡壁增厚,炎性细胞浸润;肺含水量增加;MDA和H_(2)O_(2)含量升高,GSH含量、T-SOD和CAT活力下降,IL-1β和VEGF升高(P<0.01,P<0.05)。与HG组比较,HPE组与HYP各剂量组大鼠肺组织炎性细胞浸润减轻,肺泡腔扩张并趋于正常;肺含水量降低;GSH含量、T-SOD和CAT活力升高,MDA、H_(2)O_(2)、IL-1β、IL-6、VEGF和TNF-α含量降低(P<0.01,P<0.05)。结论HPE对HAPE大鼠具有一定的保护作用,有望开发成为治疗HAPE的药物制剂。
Objective To study the protective effect of Hypericum Perforatum L.Extract(HPE)on rats with high-altitude pulmonary edema(HAPE).Methods Eighty Wistar rats were randomly divided into normoxia blank group(NG),hypoxia group(HG),low-dose HPE group(HPE-L,100 mg/kg),moderate-dose HPE group(HPE-M,200 mg/kg),high-dose HPE group(HPE-H,400 mg/kg),low-dose Hyperoside group(HYP-L,50 mg/kg),high-dose Hyperoside group(HYP-H,100 mg/kg)) and Dexamethasone group(DXM,4 mg/kg)with 10 rats in each group.All groups were given continuous intragastric administration for 7 d,and at the 4^(th) d of administration,all groups except NG group were placed in a simulated 7500 m altitude plateau environment for 72 h to establish rat models of HAPE.Pathological changes of lung tissues of rats were observed.The lung water content,contents of malondialdehyde(MDA),hydrogen peroxide(H_(2)O_(2))and reduced glutathione(GSH),vitality abilities of total superoxide dismutase(T-SOD)and catalase(CAT)and contents of inflammatory factors[interleukin-1β(IL-1β),interleukin-6(IL-6),vascular endothelial growth factor(VEGF)and tumor necrosis factor-α(TNF-α)]were detected.Results Compared with those in NG,rats in HG group had congestion and edema in pulmonary tissues,thickened alveolar wall,inflammatory cell infiltration and increased lung water content;contents of MDA and H_(2)O_(2) were significantly increased,while GSH content,vitality abilities of T-SOD and CAT were significantly decreased,and contents of IL-1βand VEGF were significantly increased in HG group(P<0.01,P<0.05).Compared with those in HG group,degrees of inflammatory cell infiltration were significantly alleviated,and alveolar cavity was dilated and tended to be normal in lung tissues;lung water contents were significantly decreased;GSH contents,vitality abilities of T-SOD and CAT were significantly increased,while contents of MDA,H_(2)O_(2),IL-1β,IL-6,VEGF and TNF-αwere significantly decreased in HPE groups and HYP groups(P<0.01,P<0.05).Conclusion HPE has a certain protective effect on rat
