摘要
本试验用水性佐剂副猪嗜血杆菌(HPS)灭活疫苗作为稀释液稀释猪瘟(CSF)活疫苗,通过测定混合疫苗中的猪瘟病毒(CSFV)含量,评价HPS灭活疫苗的稀释效果。结果显示,HPS灭活疫苗作为稀释液稀释CSF活疫苗在2 h内对猪瘟病毒含量无影响。用HPS、CSF混合疫苗免疫21~28日龄仔猪10头,42~49日龄进行HPS灭活苗二免,56~63日龄时对其中5头进行HPS 4型强毒株攻毒,5头进行HPS 5型强毒株攻毒,同时设HPS灭活疫苗免疫攻毒组、HPS 4攻毒对照组和HPS 5攻毒对照组,通过HPS免疫攻毒试验评价混合疫苗HPS部分的免疫效果,结果显示,混合疫苗免疫组与HPS灭活疫苗免疫组相比,HPS部分的效力无明显差异。用HPS、CSF混合疫苗免疫21~28日龄仔猪5头,同时设CSF活疫苗对照组,通过测定免后CSFV ELISA抗体和中和抗体评价混合疫苗CSF部分的免疫效果,结果显示,从ELISA抗体水平看,混合疫苗免疫组CSF部分的效力略优于CSF活疫苗免疫组;从中和抗体水平看,两组疫苗差异不显著。结果表明,用HPS灭活疫苗作为稀释液稀释CSF活疫苗,一次注射同时接种这2种疫苗的方案可行,为临床中简化这2种疫苗的免疫程序提供参考依据。
This study utilized Haemophilus parasuis(HPS) inactivated vaccine with aqueous adjuvant was used as a diluent for classical swine fever(CSF) live vaccine to prepare of the mixed vaccine, and evaluated the dilution effect of HPS inactivated vaccine by determining the classical swine fever virus(CSFV) content in the mixed vaccine. The result showed that HPS inactivated vaccine as the diluent for CSF live vaccine had no effect on the content of CSFV within 2 h. 10 piglets of 21 to 28 days old were immunized with the mixed vaccine of HPS inactivated vaccine and CSF live vaccine for the first time, and the secondary immunization was carried out with only HPS inactivated vaccine at the age of 42 to 49 days. Then, 5 piglets were challenged with the virulent strain of HPS serotype 4,and the other 5 piglets were challenged with the virulent strain of HPS serotype 5. At the same time, 3 control groups of HPS inactivated vaccine vaccinated-challenged, no-vaccinated but HPS serotype 4 challenged and no-vaccinated but HPS serotype 5 challenged were set up. The immunoprotective efficacy of the mixed vaccine against HPS infection was evaluated by HPS immunization and challenge tests. The result revealed that there was no significant difference between the mixed vaccine group and the HPS inactivated vaccine group in the protective efficacy against HPS infection. 5 piglets at the age of 21-28 days were immunized with the mixed vaccine, and the control group was set up with the CSF live vaccine at the same time. The immunizing effect of the part of CSF of the mixed vaccine was evaluated by measuring CSFV ELISA antibody and neutralizing antibody after immunizing pigs. In terms of ELISA antibody level,the immunizing effect of CSF part of the mixed vaccine group was slightly better than that of the live vaccine group. There was no significant difference between the two groups based on the neutralization antibody titers.
