参芪扶正汤对盆腔炎性疾病后遗症小鼠上生殖道细胞自噬水平的影响 被引量:2

Effect of Shenqi Fuzheng Decoction on Autophagy Level of Upper Reproductive Tract Cells in Mice with Sequelae of Pelvic Inflammatory Disease

摘要 目的:研究参芪扶正汤不同途径给药对盆腔炎性疾病后遗症(SPID)小鼠模型的治疗作用,并探究参芪扶正汤对SPID小鼠模型上生殖道组织中肿瘤坏死因子α(TNF-α)及自噬相关蛋白[Beclin-1、微管相关轻链蛋白3(LC3)]的影响,探讨潜在治疗机制。方法:选取60只SPF级雌性BALB/c小鼠,7~8周龄,随机分为空白对照组、模型组、西药组、中药灌胃组、中药灌肠组、中药联合组。造模成功后中药组分别予中药灌胃、灌肠及灌胃联合灌肠,空白对照组及模型组予羧甲基纤维素钠灌胃、西药组予阿司匹林肠溶片溶液灌胃。治疗15 d后评估各组小鼠脸谱疼痛评分(MGS),蛋白质免疫印迹法(WB)测定上生殖道组织Beclin-1、LC3-I及LC3-II的表达水平,免疫组化检测上生殖道不同部位组织Beclin-1、LC3的蛋白定位表达。结果:模型组小鼠MGS,TNF-α、Beclin-1及LC3-II表达均高于空白对照组。中药灌胃、灌肠、联合组小鼠MGS、TNF-α均低于模型组(P<0.05)。中药灌胃、灌肠、联合组Beclin-1、LC3-I、LC3-II水平均明显高于模型组(P<0.05)。结论:参芪扶正汤可以改善SPID慢性盆腔痛的症状、降低小鼠上生殖道组织炎症因子表达、促进细胞自噬;调节细胞自噬可能是参芪扶正汤治疗SPID的潜在机制;口服联合灌肠给药较单一途径给药而言并不能明显提高治疗SPID的临床疗效。 Objective:To study the therapeutic effect of Shenqi Fuzheng Decoction on the mouse model with sequelae of pelvic inflammatory disease(SPID),to explore the effect of Shenqi Fuzheng Decoction on tumor necrosis factorα(TNF-α)in the upper reproductive tract of SPID mouse model and autophagy related proteins(Beclin-1)and microtubule associated light chain protein 3(LC3),and to explore the potential therapeutic mechanism.Methods:60 SPF female BALB/c mice aged 7 to 8 weeks were randomly divided into control group,model group,western medicine group,Chinese medicine gavage group,Chinese medicine enema group and Chinese medicine combination group.After successful modeling,the Chinese medicine groups were given gavage,enema and combined gavage-enema respectively,while the control group and model group were given gavage with sodium carboxymethylcellulose,and the western medicine group with aspirin enteric coated tablet solution.After 15 days of treatment,mouse grimace scale(MGS)of mice in each group was evaluated,the expression levels of Beclin-1,LC3-I and LC3-II in upper genital tract were measured by Western blot(WB),and the protein localization expression of Beclin-1 and LC3 in different parts of upper genital tract was detected by immunohistochemistry.Results:MGS,TNF-α,Beclin-1 and LC3-II expressions in the model group were higher than those in the control group.MGS and TNF-αin mice of the three Chinese medicine groups were lower than those in the model group(P<0.05).Beclin-1,LC3-I and LC3-II in Chinese medicine groups were significantly higher than those in the model group(P<0.05).Conclusion:Shenqi Fuzheng Decoction can improve the symptoms of chronic pelvic pain in SPID,reduce the expression of inflammatory factors in mouse upper reproductive tract and promote autophagy;Regulating autophagy may be the potential mechanism of Shenqi Fuzheng Decoction in the treatment of SPID;Compared with single route administration,oral administration combined with enema can not significantly improve the clinical efficacy.
