摘要
为了解貉源大肠杆菌的耐药性及Ⅰ型整合子的携带情况,试验采用药敏试验对150株貉源大肠杆菌进行耐药性检测,采用PCR方法对其进行Ⅰ型整合酶intⅠ1、intⅠ2和intⅠ3和Ⅰ型整合子的检测。结果表明:150株貉源大肠杆菌对氨苄西林(AMP)、阿莫西林(AML)、头孢噻吩(KF)和四环素(TE)的耐药率均达到了80%以上;对亚胺培南(IPM)、庆大霉素(CN)、阿米卡星(AK)、诺氟沙星(NOR)、环丙沙星(CIP)、氧氟沙星(OFX)和多黏菌素B(PB)的敏感率均达到了70%以上;均表现出不同程度的耐药性,耐药种数为1~19种,多重耐药情况达到92.67%(139/150),其中,6重耐药菌株的比例最大,为12.00%(18/150)。整合酶基因intⅠ1的阳性样本数量为108株,检出率为72.00%(108/150);整合酶基因intⅠ2的阳性样本数量为14株,检出率为9.33%(14/150);均未检出整合酶基因intⅠ3。Ⅰ型整合子的阳性菌株数量为51株,检出率为34.00%(51/150),且存在1个菌株中含有最多3个基因盒的情况。Ⅰ型整合子基因盒包括aadA2、dfrA5、dfrA1-aadA1、dfrA17-aadA5、lnu(F)-aadA和空整合子,除空整合子以外,占比最大的基因盒为dfrA1-aadA1;各基因盒片段大小分别为1 074 bp(3/150)、1 097 bp(1/150)、1 645 bp(9/150)、1 727/1 729 bp(8/150)、2 002/2 014 bp(6/150)和153 bp(43/150)。说明Ⅰ型整合子在细菌耐药的传播方面起着十分重要的作用。
In order to understand the drug resistance of E. coli from raccoon dogs and the carrying situation of type Ⅰ integron, the drug resistance of 150 strains of E.coli strains from raccoon dogs was detected by drug sensitivity tests. PCR was used to detect the sequence of type Ⅰ integrase intⅠ1, intⅠ2, intⅠ3 and type Ⅰ integron. The results showed that the drug resistance rates of 150 E.coli strains to AMP, AML, KF and TE were more than 80%. The drug susceptible rates to IPM, CN, AK, NOR, CIP, OFX and PB were all over 70%. The number of drug-resistant strains ranged from 1 to 19, and the number of multiple drug-resistant strains reached 92.67%(139/150). Among them, the proportion of six drug-resistant strains was the largest, accounting for 12.00%(18/150). The number of positive samples of integrase gene intI1 was 108 strains, with a detection rate of 72.00%(108/150). The number of positive samples of integrase gene intI2 gene was 14 strains, with a detection rate of 9.33%(14/150). The integrase gene intI3 was not detected. The number of type Ⅰ integron positive strains was 51, with a detection rate of 34.00%(51/150), and one strain contained up to 3 gene boxes. The type Ⅰ integron loci included aadA2, dfrA5, dfrA1-aadA1, drfA17-aadA5, lnu(F)-aadA and empty integrators. In addition to empty integrators, dfrA1-aadA1 accounted for the largest proportion. The size of each loci was 1 074 bp(3/150), 1 097 bp(1/150), 1 645 bp(9/150), 1 727/1 729 bp(8/150), 2 002/2 014 bp(6/150) and 153 bp(43/150), respectively. These results indicated that type Ⅰ integron played an important role in the transmission of bacterial drug resistance.
