光叶子花及紫茉莉科植物叶绿体基因组序列的比较分析 被引量:4

摘要 对光叶子花叶绿体基因组特征进行分析,探究其与近缘种之间的系统发育关系。本研究通过NCBI数据库,下载已公布的石竹目(Caryophyllaceae)12个种的叶绿体基因组序列,以秘鲁叶子花叶绿体基因组作为参考,通过Genious中的MAFFT软件进行序列比对,并利用生物信息学手段进行特征分析。结果显示:光叶子花叶绿体基因组长度为154763 bp,LSC区长度为85921 bp,SSC区长为18018 bp,一对IR区均长25412 bp,呈典型的四分体结构。注释基因有131个,蛋白编码基因86个,tRNA基因37个还有8个rRNA基因,平均GC含量为36.4%。利用MISA软件在紫茉莉科9个种的叶绿体基因组中共检测到605个SSR,重复类型中以单核苷酸最多582个,占总SSR的96.2%。重复基元最多的是A/T,572个,占比94.5%。紫茉莉科不同物种的叶绿体基因组IR边界区的基因类型和基因分布情况存在一定差异,但相对比较保守。紫茉莉科9个种叶绿体基因组之间检测到6个高变区,rps16-trnQ、rps4-trnL、ndhC-trnV、rps3-rpl22、ndhF-rpl32和ycf1。系统发育分析表明,光叶子花和叶子花在亲缘关系上关系最近。 To analyze the characteristics of chloroplast genome of Bougainvillea glabra and explore its phylogenetic relationship with related species.The chloroplast genome sequences of 12 species of Caryophyllaceae were downloaded from NCBI database,and the chloroplast genome of B.peruviana was used as reference.The sequences alignment was performed by MAFFT software in Genious and the characteristics were analyzed by bioinformatics methods.The results showed that the chloroplast genome length was 154763 bp,the LSC region length was 85921 bp,the SSC region length was 18018 bp,and a pair of IR regions were 25412 bp,which showed a typical tetrad structure.Annotated genes has 131,86 protein coding genes,37 tRNA genes and 8 rRNA genes,and the average GC content was 36.4%.A tolal of 605 SSR loci were detected in the chloroplast genomes of 9 species of Nyctaginaceae by MISA software,of which 582 were single nucleotide,accounting for 96.2%of the total SSRs.The largest number of repeat primitives were A/T,572,accounting for 94.5%.There were some differences in gene types and gene distribution in IR boundary region of chloroplast genome among different species of Nyctaginaceae,but they were relatively conservative.Six hypervariable regions,rps16-trnQ、rps4-trnL、ndhC-trnV、rps3-rpl22、ndhF-rpl32 and ycf1,were detected among the 9 chloroplast genomes.Phylogenetic analysis showed that B.glabra and B.spectabilis were closely related in genetic relationship.
