马鞭草总苷对大鼠慢性前列腺炎/骨盆疼痛综合征的改善作用及机制研究 被引量:6

Improving Effect and Mechanism of Total Glycosides of Verbena officinalis L.for Chronic Prostatitis/Pelvic Pain Syndrome in Rats

摘要 【目的】探讨马鞭草总苷对慢性前列腺炎/骨盆疼痛综合征(CP/CPPS)模型大鼠的治疗机制。【方法】将40只SPF级雄性SD大鼠随机分为空白对照组、模型对照组和马鞭草总苷低、中、高剂量组,每组8只。除空白对照组外,其余各组均采用同源大鼠前列腺蛋白提纯液辅以完全弗氏佐剂诱导CP/CPPS大鼠模型。马鞭草总苷低、中、高剂量组依次给予30、60、120 mg·kg^(-1)的马鞭草总苷灌胃,空白对照组和模型对照组则给予等体积的生理盐水灌胃,连续22 d,每日1次。末次给药后的第2天,用Von Frey纤维丝测痛仪测定机械痛阈值。处死大鼠,称量体质量及前列腺湿质量,计算前列腺指数。采用苏木素-伊红(HE)染色法观察前列腺组织病理变化;酶联免疫吸附分析(ELISA)检测前列腺组织白细胞介素1β(IL-1β)、白细胞介素8(IL-8)及肿瘤坏死因子α(TNF-α)含量;蛋白免疫印迹(Western Blot)法检测前列腺组织总p38丝裂原活化蛋白激酶(p38MAPK)、磷酸化p38MAPK(p-p38MAPK)及基质金属蛋白酶9(MMP-9)的蛋白相对表达量;流式细胞术检测外周血T辅助淋巴细胞(Th)1及Th2百分比。【结果】与模型对照组比较,马鞭草总苷各剂量组前列腺纤维增生及炎症细胞浸润明显改善,前列腺指数均显著降低,机械痛阈值均显著增加,TNF-α、IL-1β及IL-8含量均显著降低,p-p38MAPK、MMP-9蛋白相对表达量及p-p38MAPK/p38MAPK比值均显著降低,Th1淋巴细胞百分比及Th1/Th2比值均显著降低,Th2淋巴细胞百分比显著升高(P<0.05或P<0.01)。马鞭草总苷各剂量组组间比较,除Th2淋巴细胞百分比外,其余指标均有显著的统计学差异(P<0.05或P<0.01)。【结论】马鞭草总苷对CP/CPPS模型大鼠有抗炎、镇痛作用,其机制可能与下调p38 MAPK信号通路相关调控蛋白和调节Th1/Th2细胞亚群比例有关。 Objective To investigate the treatment mechanism of total glucosides of Verbena officinalis L.(TGV)for model rats with chronic prostatitis/pelvic pain syndrome(CP/CPPS).Methods Forty SPF male SD rats were randomly divided into blank control group,model control group and TGV low-,medium-and high-dose groups,with 8 rats in each group.Except the blank control group,CP/CPPS model was induced by homologous rat prostate protein extract supplemented with complete Freund’s adjuvant in other groups.TGV low-,medium-and highdose groups were intragastrically given 30,60 and 120 mg·kg^(-1) of TGV,respectively,while the blank control group and model group were intragastrically given equal volume of normal saline.The administrations performed once a day for 22 consecutive days.On the second day after the last administration,the mechanical pain threshold was measured with the Von Frey pain threshold detector.After the rats were sacrificed,body mass and wet mass of prostate were weighed,and prostate index was calculated.The pathological changes of prostate tissues were observed by hematoxylin-eosin(HE)staining;the contents of interleukin 1β(IL-1β),interleukin 8(IL-8)and tumor necrosis factorα(TNF-α)in prostate tissues were determined by enzyme-linked immunosorbent assay(ELISA);the relative protein expression levels of p38 mitogen-activated protein kinase(p38MAPK),phosphorylated p38MAPK(p-p38MAPK)and matrix metalloproteinase 9(MMP-9)in prostate tissues were detected by Western Blot method;the percentage of T helper lymphocyte(Th)1 and Th2 in peripheral blood were detected by flow cytometry.Results Compared with the model control group,the prostatic fiber hyperplasia and inflammatory cell infiltration were significantly improved,the prostatic index was significantly decreased,the mechanical pain threshold was significantly increased,and the contents of TNF-α,IL-1βand IL-8 were significantly decreased,the relative protein expression levels of p-p38MAPK and MMP-9 and the ratio of p-p38MAPK/p38MAPK were significantly decrease
