摘要
为探讨Cas9基因编辑技术应用在小型巴马猪时,在麻醉状态下对其生命体征的影响和重要脏器组织学变化,为基因编辑巴马猪在麻醉手术中的应用提供基础生理数据。使用诱导麻醉配合呼吸麻醉对20头野生型巴马猪和24头Cas9基因编辑型巴马猪进行麻醉,监测麻醉实验动物的呼吸频率、心率、体温和血压等生命体征,比较麻醉后野生型巴马猪和基因编辑型巴马猪之间生命体征和重要脏器组织学差异。结果表明,在排除月份差异的情况下,野生型巴马猪和Cas9基因编辑型巴马猪的体长和体重无明显差异(P>0.05),在麻醉状态下,二者呼吸频率、心率、体温和血压均无显著性差异(P>0.05);与野生型相比Cas9基因编辑型巴马猪的肺脏、心脏、肝脏及肾脏等重要脏器的组织病理学不存在明显变化。研究表明,Cas9基因编辑技术对巴马猪的生命体征无影响,对重要组织器官组织学上影响不明显。
To investigate the basic data of vital signs and the important organs’ histopathologic changes of Bama minipigs(which have used Cas9 gene editing techniques) under anesthesia condition, 20 healthy wild type and 24 healthy Cas9 gene edited pigs were used to measure the respiratory rate, heart rate, temperature, systolic pressure, diastolic pressure and arterial blood pressure by ECG monitoring and histologic examination.Paraffin sections of lung, heart, liver and kidney tissue stained by haematoxylin and eosin were examed after euthanisation.Compared with wild type Bama minipigs, there’s no significant differences in respiratory frequency, heart rate, temperature and blood pressure in Cas-9 gene edited Bama minipigs(P>0.05).Also there’s no significant differences in histopathologic changes of lung, heart, liver and kidney between the two types of pigs.The CRISPR/Cas9 gene editing technique has no effect on vital signs and histopathology of Bama minipigs under anesthesia.
