痰湿内阻慢性失眠大鼠模型建立及半夏秫米汤基于HPA轴、炎症因子的治疗机制 被引量:15

摘要 目的探讨痰湿内阻慢性失眠大鼠模型建立及半夏秫米汤基于HPA轴、炎症因子的治疗机制。方法将48只Wistar大鼠随机分为空白组、痰湿内阻慢性失眠组及半夏秫米汤低、中、高剂量组和地西泮组,每组8只。高脂饲料+猪油灌胃7周后,继以单平台水环境睡眠剥夺法3周复制痰湿内阻慢性失眠模型。空白组、痰湿内阻慢性失眠组予1 mL/100 g生理盐水灌胃,半夏秫米汤低、中、高剂量组按4.69、9.38、18.75 g/kg灌胃,地西泮组按0.75 mg/kg地西泮水溶液灌胃,每日1次,连续14 d。给药14 d后采用ELISA法检测HPA轴关键分子及炎症因子表达。结果与空白组相比,痰湿内阻慢性失眠模型组血清TG、LDL-C升高,HDL-C降低(P均<0.01);睡眠潜伏期延长,睡眠持续时间缩短(P<0.05);与痰湿内阻慢性失眠模型组相比,半夏秫米汤各给药组血清ACTH、CORT、CRH、GABA、NE、DA含量降低,下丘脑IL-1β、TNF-α降低,下丘脑IL-4、IL-10含量升高(P均<0.05)。结论高脂饲料+单平台水环境法复制痰湿内阻慢性失眠大鼠模型可行,半夏秫米汤干预痰湿内阻慢性失眠的作用机制是通过抑制亢进的HPA轴,调Shumi Decoction intervening chronic insomnia is achieved through inhibiting the hyperactive HPA axis and regulating the expression levels of inflammatory factors,GABA,NE,and DA. Objective To explore the establishment of rat models of chronic insomnia duo to phlegm-dampness inter⁃nal obstruction and the therapeutic mechanism of Banxia Shumi decoction based on HPA axis and inflammatory factors.Methods Forty-eight Wistar rats were randomly divided into the blank group,chronic insomnia duo to phlegm-dampness internal obstruction group,low-dose,medium-dose and high-dose Banxia Shumi decoction groups,and diazepam group,with 8 rats in each group.The rat models of chronic insomnia duo to phlegm-dampness internal obstruction were estab⁃lished by sleep deprivation with single platform water for 3 weeks after 7 weeks of high fat diet plus lard gavage.The nor⁃mal saline(1 mL/100 g)was given by gavage to the rats in the blank group and chronic insomnia duo to phlegm-dampness internal obstruction group;4.69,9.38,and 18.75 g/kg Banxia Shumi decoction was given by gavage to the rats in the low-dose,medium-dose and high-dose groups,respectively,and the rats in the diazepam group were given 0.75 mg/kg di⁃azepam aqueous solution,once a day,for consecutive 14 days.The expression of HPA axis key molecules and inflammato⁃ry factors was detected by ELISA after 14 days of administration.Results Compared with the blank group,serum TG and LDL-C increased and HDL-C decreased(both P<0.01),sleep latency was prolonged and sleep duration was short⁃ened in the chronic insomnia duo to phlegm-dampness internal obstruction group(both P<0.01);compared the chronic in⁃somnia duo to phlegm-dampness internal obstruction group,the content of ACTH,CORT,CRH,GABA,NE and DA in serum,IL-1βand TNF-αin hypothalamus decreased,IL-4 and IL-10 in hypothalamus increased in all Banxia Shumi de⁃coction groups(all P<0.05).Conclusions High fat diet+single platform water environment method is a feasible meth⁃od to replicate the rat model of chronic insomnia duo to phlegm-dampness internal obstruction.The mechanism of Banxia Shumi Decoction intervening chronic insomnia is achieved through inhibiting the hyperactive HPA axis a
