摘要
【目的】为解析不同马铃薯组织中糖苷生物碱的积累对人类使用及食品加工是否产生影响,研究不同储藏条件下马铃薯不同组织器官中的糖苷生物碱积累水平。【方法】用高效液相色谱—串联质谱法检测1-76和6-36(紫皮)、5-36和6-1(白皮)4种马铃薯资源在光照和地窖储藏60 d后块茎和芽2种组织中的糖苷生物碱含量变化情况;并利用实时荧光RCR对糖苷生物碱生物合成相关基因的表达量进行分析。【结果】糖苷生物碱检测显示:不同条件储藏后4种马铃薯芽中的糖苷生物碱含量显著高于块茎中的糖苷生物碱含量,马铃薯芽及马铃薯6-36(0.272 mg/g)和5-36(3.360 mg/g)块茎中的糖苷生物碱含量均已超出安全食用标准(0.2 mg/g,鲜质量),但是否能安全食用及食品加工需要更多试验验证;糖苷生物碱生物合成相关基因表达量检测显示:不同马铃薯资源糖苷生物碱生物合成途径中部分基因(StCAS,StSSR2,StGAME4,StSGT1,StSGT2)的表达量与糖苷生物碱含量关系密切,其他基因在马铃薯中的表达具有一定的组织特异性。【结论】本试验通过简单、快速地检测马铃薯块茎、芽中的糖苷生物碱含量,发现在储藏后马铃薯资源(1-76、6-1)块茎中的糖苷生物碱含量虽未超出安全标准,但不建议食用,本研究结果为人类安全食用马铃薯提供了理论参考价值。
[Objective]The present paper aimed to analyze whether the accumulation of glycoside alkaloids in different potato tissues has influence on human use and food processing,the accumulation levels of glycoside alkaloids in different potato tissues and organs under different storage conditions were summarized.[Method]In the present study,the contents of glucoside alkaloids in tuber and bud tissues of potato resources 1-76 and 6-36(purple bark),5-36 and 6-1(white bark)were determined by HIGH performance liquid chromatography-tandem mass spectrometry after 60 days of storage in light and cellars.Real-time fluorescence RCR was used to analyze the gene expression of glycoside alkaloid biosynthesis.[Result]The detection of glycoalkaloids showed that the content of glycoalkaloids in four kinds of potato sprouts was significantly higher than that in tubers after storage under different conditions,the content of glycoalkaloids in tubers exceeded the safe edible standard(0.2 mg/g,fresh quality),but whether it was safe to eat and food processing needs more experimental verification;The detection in the expression of glycoalkaloid biosynthesis-related genes showed that the expression levels of some genes(StCAS,StSSR2,StGAME4,StSGTl,StSGT2)in the glycoalkaloid biosynthesis pathway of different potato resources were closely related to the content of glycoalkaloids,and the expression of other genes in potato had certain tissue specificity.[Conclusion]By simply and quickly detecting the content of glycoalkaloids in potato tubers and sprouts,this test found that the content of glycoalkaloids in tubers of potato resources(1-76,6-1)after storage did not exceed the safety standard,but it was not recommended.The results of this study provided a theoretical reference value for human safe consumption of potatoes.
