摘要
为深入了解毛竹中的热激蛋白(heat shock protein,HSP)及其特性,借助生物信息学工具对毛竹HSP90家族基因进行鉴定,分析其理化性质、结构特征、系统进化关系以及组织特异性表达模式。结果表明,毛竹基因组中含11个HSP90家族基因,分布于9条scaffold上,外显子5~20个不等;系统进化关系分析显示毛竹11个HSP90家族基因分属于3组,结构特征相似的成员分属于同一组;共线性分析表明毛竹中仅有1个HSP90基因(PH02Gene09089)与水稻的HSP90基因不存在共线性关系;亚细胞定位结果显示毛竹HSP90蛋白定位于细胞质或内质网中;HSP90家族成员启动子上分布有丰富的与激素、光照、及逆境相关的顺式作用元件;已发表的转录组数据及RT-PCR结果表明,HSP90家族各基因在不同组织中的表达模式不同。本研究结果加深了对毛竹HSP90基因家族的了解,可为研究该家族基因的生物学功能及后续利用提供参考。
In order to understand the HSP90 gene family in Moso bamboo and their characteristics,this study identified the HSP90 family genes,and analyzed their physical and chemical properties,structural characteristics,phylogenetic relationships and tissue-specific expression patterns by bioinformatics tools.The results showed that 11 HSP90 genes were identified and were located on nine scaffolds.The HSP90 gene family contained 5~20 exons.Phylogenetic analysis divided the Moso bamboo HSP90 genes into three groups.The structural characteristics of HSP90 members in the same group were similar but varied among different groups.Synteny analysis showed that only one moso bamboo HSP90 gene,PH02Gene09089,was conservative,which was not collinear with any HSP90 gene in rice.The subcellular localization analysis indicated that HSP90 targeted the cytoplasm or endoplasmic reticulum.The promoter analysis showed that the promoters of HSP90 genes contained hormone,light,and stress-related cis-elements.Moreover,the HSP90 gene family showed different expression profile in different tissues by analyzing published RNA-Seq data and RT-PCR.These findings improve the understanding of Moso bamboo HSP90 family genes and provide theoretical basis for deeper studies of their biological functions in plant response to abiotic stress.
