摘要
铁死亡是铁依赖性的,不同于细胞凋亡、细胞坏死和细胞自噬的一种新型的细胞程序性死亡方式,其特征为铁依赖性脂质过氧化物积累伴有氧化还原酶功能的下降,特别是由于谷胱甘肽过氧化物酶4(GPX4)活性降低导致脂质过氧化物的堆积引起铁死亡。铁死亡作为细胞死亡的一种方式参与机体正常生理和病理调节过程。铁死亡可能损害葡萄糖刺激胰岛β细胞分泌胰岛素而参与糖尿病发病机制。近年来的研究显示,铁死亡在糖尿病以及糖尿病各种并发症的发生、发展中发挥重要调控作用。本文针对铁死亡的调控机制及其在糖尿病并发症中的研究进展进行综述。
Ferroptosis is a new and iron-dependent programmed cell death,which differs from apoptosis,cell necrosis and autophagy.Ferroptosis is characterized by iron-dependent lipid peroxideaccumulation accompanied by inadequate redox enzymes,especially the accumulation of lipid peroxides caused by loss of glutathione peroxidase 4(GPX4).Ferroptosis,as a type of cell death,is invovled in both physiologic and pathogenic processes.Ferroptosis may impair glucose-stimulated insulin secretion in isletβcells and participate in the pathogenesis of diabetes.Recent studies have shown that ferroptosis plays a crucial role in the development of diabetes mellitus and its associated complications.In this review,we summarize recent advances in the mechanisms of ferroptosis and its role in the development of diabetic complications.
