摘要
目的研究菌种保藏管法和冷冻真空干燥保藏法中对金黄色葡萄球菌抗力的影响。方法菌种分别用以下两种保存方法进行保存:冷冻真空干燥保藏法4℃保存和菌种保藏管法-80℃保存。测试速干手消毒液对不同菌种保藏方法的金黄色葡萄球菌的杀菌率。结果随着保存时间延长,菌种保藏管法的金黄色葡萄球菌抗力下降明显,而冷冻真空干燥保藏法的金黄色葡萄球菌抗力未见明显变化。采用菌种保藏管法保存6个月、1年、3年,消毒液对金黄色葡萄球菌的杀菌率分别为99.076%、99.193%、99.968%,差异有统计学意义(P<0.05);采用冷冻真空干燥保藏法保存6个月、1年、3年,消毒液对金黄色葡萄球菌杀菌率分别为99.025%、99.039%、99.063%,差异无统计学意义(P>0.05)。结论随着菌种保存时间延长,冷冻真空干燥保藏法对金黄色葡萄球菌抗力影响较小,保存效果优于菌种保藏管法。
Objective To study the influence by the microbial preservation tube and vacuum freeze-drying microbial preservation of microbial strain preservation on the resistance variety of Staphylococcus aureus.Methods The strains were preserved by the following two preservation methods:vacuum freeze-drying microbial preservation method at 4℃and microbial preservation tube method at-80℃.The suspension quantitative germicidal test was used to observe the resistance of Staphylococcus aureus with the two different methods of microbial strain preservation.Results Compared with Staphylococcus aureus on the vacuum freeze-drying microbial preservation,the resistance variety of the microbial preservation tube was significantly decreased as time goes on.The bactericidal rates of disinfectants against Staphylococcus aureus were 99.076%,99.193%and 99.968%,after the microbial preservation tube method was used for 6 months,1 year and 3 years.Differences were statistical significance(P<0.05).The bactericidal rates of disinfectants against Staphylococcus aureus were 99.025%,99.039%and 99.068%,after the vacuum freeze-drying microbial preservation was used for 6 months,1 year and 3 years,but there were no statistical difference(P>0.05).Conclusion It was relatively little impact on the Staphylococcus aureus′resistance variety of the vacuum freeze-drying microbial preservation as time goes on.The effect of the vacuum freeze-drying preservation is better than that of microbial preservation tube.
