摘要
目的建立测定壮腰健肾片中原儿茶酸含量的高效液相色谱(HPLC)法。方法采用C_(18)色谱柱(4.6×250 mm,5μm;或4.6×150 L,5μm)色谱柱,乙腈-磷酸缓冲液(磷酸二氢铵11.5 g和磷酸1 mL,加水至1000 mL使溶解)(5:95)为流动相;检测波长为260nm。柱温:40℃;进样量:10μL;流速:1.0 mL·min^(-1)。结果原儿茶酸在0.01 mg·mL^(-1)~0.09 mg·mL^(-1)范围内线性良好(r=0.99998),平均回收率为98.7%,RSD=1.3%(n=9)。结论该方法简便、重复性好、准确,可作为壮腰健肾片的原儿茶酸含量质量控制。
OBJECTIVE To establish a HPLC method for the determination of protocatechuic acid in Zhuangyao Jianshen Tablets.METHODS C_(18) column(4.6×250 mm,5μm;or 4.6×150 L,5μm)was used.The mobile phase was acetonitrile-phosphoric acid buffer solution(11.5 g ammonium dihydrogen phosphate and 1 mL phosphoric acid,add water to 1000 mL to dissolve)(5∶95).The detection wavelength was 260 nm.The column temperature was 40℃.The injection volume was 10μL.The flow rate was 1.0 mL·min^(-1).RESULTS The calibration curves was linear in the concentration range of 0.01 mg·mL^(-1)~0.09 mg·mL^(-1)(r=0.99998),the average recovery was 98.7%,RSD=1.3%(n=9).CONCLUTION The method is simple,reliable and accurate,which is suitable for the quality control of protocatechuic acid in Zhuangyao Jianshen Tablets.
