摘要
目的探讨三化汤治疗中风的成分、靶点、活性、通路,为三化汤的分子作用机制研究及产品质量提升提供科学依据。方法应用网络药理学平台,分析三化汤组成药味(大黄、枳实、厚朴和羌活)在治疗中风处方中出现的频度及作用靶标,分析其治疗中风的活性成分、作用靶点及通路,探讨其分子作用机制。结果预测到核心成分24种,核心靶标18个,核心通路7条,成药性好的成分有橙皮素、丁香酸、厚朴脂素M、6′-甲氧基和厚朴酚、异厚朴酚、丁香醛和香草酸;涉及抗炎的核心靶标有VHL、NR3C1、ESR1和NFKBIA等,涉及的主要通路有白细胞介素-4和白细胞介素-13信号通路、TAK1通过磷酸化和IKKS激活的NF-κB通路、TRAF6参与NF-κB介导的激活通路、类固醇激素受体(steroid hormone receptors,SHR)的热休克蛋白90伴侣循环通路、核受体转录途径、Ub-特异性加工蛋白酶途径、细胞受体泛素相关修饰途径。结论三化汤治疗中风与抗炎保护具有密切的关系。
Objective To investigate the composition,target,activity and pathway of Sanhua Decoction in the treatment of stroke,therefore to provide a basis for exploring the molecular mechanism and exploiting the product.Methods Through the platform of network pharmacology,the frequency of Sanhua Decoction in the prescription of treating stroke,active target and active components,targets and the pathways were analyzed to explore the molecular mechanism.Results 24 crucial ingredients,18 core targets and 7 related pathways were predicted.Drug-likeness grading of these compositions included hesperitin,cedar acid,houpulin M,6′-O-methylhonokiol,isomagnolol,syringaldehyde,and vanilic acid.The core target of anti-inflammation of this decoction majorly included VHL,NR3C1,ESR1,and NFKBIA etc..The main pathways included the pathways of interleukin-4 and interleukin-13 signaling,TAK1 activating NF-κB by phosphorylation and activation of IKKS complex,TRAF6 mediating NF-κB activation,HSP90 chaperone cycle for steroid hormone receptors(SHR),nuclear receptor transcription,Ub-specific processing proteases,and SUMOylation of intracellular receptors.Conclusion Sanhua Decoction has a close relationship with anti-inflammatory protection in treating apoplexy.
