摘要
为了准确鉴定黑龙江省丹顶鹤体内的次睾吸虫并确定其分类地位,本试验采用形态学观察与PCR扩增ITS序列进行虫种鉴定。结果显示:试验中次睾吸虫与东方次睾吸虫形态学特点基本一致;该吸虫的ITS序列长度为1077 bp,其中ITS1、5.8S rDNA、ITS2的长度分别是629、160 bp和288 bp,与NCBI中东方次睾吸虫(序列号:MK482054)的同源性达到99.8%,与其他次睾吸虫的同源性最高可达到98.2%;进化分析显示,试验中的次睾吸虫与已发表的东方次睾处于1个小分支,与其他次睾属吸虫形成了1个大分支,表明了其亲缘关系较近。基于形态学和分子生物学的鉴定结果,本试验中的次睾吸虫为东方次睾吸虫,所获得的ITS序列可作为后睾科寄生虫分类的基因标记。
This study employed morphological observation and PCR amplification of ITS sequence to determine the taxonomic status of Metorchis spp.from Grus japonensis in Heilongjiang province.The results showed that the morphological characteristics of Metorchis spp.in question were identical to those of Metorchis orientalis,the length of its rDNA ITS sequence was 1077 bp,while its ITS1,5.8S rDNA and ITS2 were 629,288 bp and 160 bp,respectively.The rDNA ITS sequence of Metorchis spp.in question shared 99.8%homology with Metorchis orientalis and 98.2%homology with other Metorchis spp.Phylogenetic analysis revealed that Metorchis spp.in question and Metorchis orientalis were in a small branch,which were in a large branch with other Metorchis spp.This study thus confirmed that Metorchis spp.in question was Metorchis orientalis,and its ITS sequence could be used as a genetic marker for future investigation of other members in the family Opisthorchiidae.
