177份小麦种质资源低分子量麦谷蛋白基因型分析 被引量:1

摘要 对177份小麦种质资源的低分子量麦谷蛋白的基因型进行分析,筛选优质亲本材料并为其利用提供依据,利用文献报道的Glu-A3、Glu-B3基因座位的相关基因特异标记,通过PCR方法检测其在177份参试研究材料的分布。结果表明,扩增到目标条带的157份参试材料中,在Glu-A3位点,携带Glu-A3d基因的材料占28.7%,携带Glu-A3e基因的材料仅有云麦20号、云南778、78鉴-35等6份,占3.8%;在Glu-B3位点,携带Glu-B3基因的材料105份,占66.9%,带有Glu-B3g和Glu-B3i基因的材料各为22份,各占14.4%;带有Glu-B3c和Glu-B3h基因的材料分别为3份和14份,分别占1.9%和8.9%;携带Glu-B3j基因的1B/1R易位系50份,占31.8%。筛选出具有优质LMW-GS组合的材料共6份,其中Glu-A3d+Glu-B3g的巧家3号、墨依和中作813等3份;带有Glu-A3d+Glu-B3i组合的滇802-87、云麦33号;带有Glu-B3g+Glu-B3i组合的绵阳11号。再根据文献报道的结果,筛选出同时具有优质高、低分子量麦谷蛋白基因组合的材料4份,带有1、17+18、5+12、A3d基因组合的材料滇832-63和昆7808,带有1、17+18、5+12、B3g基因组合的楚麦9024,带有13+16、5+12、B3g基因组合的滇801,以上10份资源可作为云南小麦品质育种重点利用的材料。 In this paper,the genotypes of glutenin with low molecular weight from 177 wheat germplasm resources were analyzed to screen high quality parents and provided basis for their utilization.Specific markers related to Glu-A3 and Glu-B3 loci were used to detect their distribution in 177 samples by PCR according to report in literatures.The results showed that the germplasms contained Glu-A3d in Glu-A3 loci accounted for 28.7%in 157 materials with target band,and the materials with Glu-A3d included Yunmai 20,Yunnan 778,78 Jian-35,etc,which accounted for 3.8%.And,105 materials contained Glu-B3 in Glu-B3 loci accounted for 66.9%,while 22 materials contained Glu-B3g and Glu-B3i accounted for 14.4%.In addition,there were 3 and 14 materials contained Glu-B3c and Glu-B3h,which accounted for 1.9%and 8.9%respectively.There were 50 lines with 1 B/1 R translocation contained Glu-B3j gene,which accounted for 3.18%.Finally,a total of 6 materials with high quality LMW-GS combination were screened,and there were 3 materials including Qiaojia 3,Moyi and Zhongzuo 813 which contained the combination of Glu-A3d+Glu-B3g,while 2 materials contained the combination of Glu-A3d+Glu-B3i were Dian 802-87 and Yunmai 33.And the material Mianyang 11 contained the combination of Glu-B3g+Glu-B3i.Based on the results reported in the literature,4 materials contained the combination with characteristics of high quality and glutenin with low molecular weight were screened,and the Dian 832-63 and Kun 7808 contained the combination of 1,17+18,5+12 and A3d.And Chumai 9024 contained the combination of 1、17+18、5+12、B3g,while Dian 801 included the combination of 13+16、5+12、B3g.The 10 materials above could be used as the key materials for quality breeding of wheat in Yunnan.
