摘要
目的:采用网络药理学方法探讨麦门冬汤治疗肺纤维化的潜在作用机制。方法:通过中药分子机制的生物信息学分析工具(BATMAN-TCM)、中医药百科全书数据库(ETCM)获取麦门冬汤的化学成分和潜在作用靶点;通过治疗靶点数据库(TTD)、人类的非线性孟德尔遗传数据库(OMIM)、DRUGBANK数据库获取肺纤维化相关的作用靶点。采用Cytoscape3.6.2软件绘制单味药-成分-共有靶点网络;采用STRING数据库并结合Cytoscape3.6.2软件绘制关键靶点相互作用图;采用David 6.8数据库进行基因本体论(GO)分析与京都基因和基因组数据库(KEGG)通路分析。结果:获得麦门冬汤392个成分,2682个潜在靶点,98个肺纤维化相关靶点,麦门冬汤和肺纤维化共有靶点49个。对单味药-成分-共有靶点网络进行网络参数分析,筛选出24个麦门冬汤抗肺纤维化的主要作用靶点。GO及KEGG分析显示,麦门冬汤抗肺纤维化的主要靶点涉及119个生物学过程,28个细胞组分,27个分子功能以及12条信号通路。结论:麦门冬汤抗肺纤维化作用可能与5-羟色胺受体(5-HTR)、转化生长因子受体(TGF)等密切相关,麦门冬汤可能通过调节神经活性配体-受体相互作用通路、钙信号通路、环磷酸腺苷(cAMP)信号通路发挥抗肺纤维化作用,有待于进一步实验验证。
Objective:To explore the potential mechanism of Maimendong Decoction(麦门冬汤)in the treatment of pulmonary fibrosis by network pharmacology.Methods:The chemical composition and potential targets of Maimendong Decoction were obtained through the Bioinformatics Analysis Tool for Molecular mechanism of Traditional Chinese Medicine(BATMAN-TCM)and the Encyclopedia of Traditional Chinese Medicine(ETCM)database.The pulmonary fibrosis-related targets were obtained through Therapeutic Target Database(TTD),Online Mendelian Inheritance in Man(OMIM)and DRUGBANK database.Cytoscape 3.6.2 software was used to draw the single herb-component-shared target network.The STRING database and Cytoscape 3.6.2 software were used to draw the key target interaction map.The David 6.8 database was used to carry out Gene Ontology(GO)analysis and Kyoto Gene and Genomic Encyclopedia(KEGG)pathway analysis.Results:Total 392 components and 2682 potential targets of Maimendong Decoction,98 pulmonary fibrosis-related targets were obtained.There were total 49 common targets of Maimendong Decoction and pulmonary fibrosis.By analyzing the network parameters of single drug-component-common target network,24 main targets of Maimendong Decoction against pulmonary fibrosis were screened out.GO and KEGG analysis showed that the key targets involved 119 biological processes and 28 cells components,27 molecular functions and 12 signal pathways.Conclusions:The anti-pulmonary fibrosis effect of Maimendong Decoction may be closely related to serotonin receptor(5-HTR)and transforming growth factor receptor(TGF),etc.Maimendong Decoction may play an anti-pulmonary fibrosis role by regulating neuroactive ligand-receptor interaction pathway,calcium signal pathway and cAMP signal pathway,which needs to be further verified by experiments.
