基于Nrf2/ARE信号通路探讨清肺通络方对肺炎支原体感染小鼠肺氧化损伤的影响 被引量:7

Effects of Qingfei Tongluo Recipe on lung oxidative damage in mycoplasma pneumoniae infected mice based on Nrf2/ARE signaling pathway

摘要 目的:基于核因子E2相关因子2/抗氧化反应元件(Nrf2/ARE)信号通路探讨清肺通络方对肺炎支原体(MP)感染小鼠肺氧化损伤的作用机制。方法:将50只BALB/c幼鼠随机分成正常组,模型组,清肺通络方低、中、高剂量组,每组10只。除正常组外,其余组幼鼠通过鼻滴入1×10^(7)CCU/mL MP菌液,每只0.1 mL,连续3d建立肺炎支原体肺炎(MPP)模型后,清肺通络方低、中、高剂量组分别以清肺通络方0.125、0.25、0.5 mL/20 g灌胃,1次/d,连续治疗5 d。正常组和模型组分别以0.9%氯化钠溶液0.25 mL/20 g灌胃。实验结束第2天进行取材,HE染色检测肺组织病理改变,Western Blot检测肺组织Nrf2、胞浆伴侣分子(Keap1)、血红素加氧酶-1(HO-1)及醌氧化还原酶(NQO1)的表达情况。结果:与模型组比较,HE染色显示清肺通络方各剂量组肺组织结构明显改善,病理评分降低,其高剂量组改善最为显著(P<0.05)。模型组小鼠肺组织中Nrf2、Keap1、HO-1、NQO1表达水平较空白组显著降低(P<0.05,P<0.01);与模型组比较,清肺通络方中、高剂量组Nrf2、Keap1、HO-1、NQO1表达水平显著上调(P<0.05);与清肺通络方低剂量组比较,清肺通络方高剂量组Nrf2、Keap1、HO-1、NQO1表达水平显著上调(P<0.05);清肺通络方高剂量组Keap1表达水平较清肺通络方中剂量组显著上调(P<0.05)。结论:清肺通络方能保护MP感染后引起的氧化应激,减轻肺损伤,其机制可能是激活Nrf2/ARE信号通路实现对肺氧化损伤的保护作用。 Objective:To explore the mechanism of Qingfei Tongluo Recipe on lung oxidative damage in mice infected with mycoplasma pneumoniae(MP)based on Nrf2/ARE signaling pathway.Methods:A total of 50 young BALB/c rats were randomly divided into normal group,model group,Qingfei Tongluo Recipe low,middle,high dose group,10 mice in each group.Except for the normal group,the young rats in the other groups were instilled with 1×10^(7)CCU/mL MP bacteria solution by nasal drip.After the model of mycoplasma pneumoniae pneumonia(MPP)was established for 3 days,the low,middle,high dose groups were treated with Qingfei Tongluo Recipe 0.125,0.25,0.5 mL/20 g,once a day for 5 days.The normal group and model group were intragastrically perfused with normal saline 0.25 mL/20 g.The pathological changes of lung tissue were detected by HE staining.The expression of nuclear factor E2 related factor 2(Nrf2),cytoplasmic chaperone molecule(Keap1),heme oxygenase-1(HO-1)and quinone oxidoreductase(NQO1)were detected by Western Blot.Results:Compared with the model group,HE staining showed that the lung tissue structure of each dose group of Qingfei Tongluo Recipe was significantly improved,and the pathological score was decreased,especially in the high-dose group(P<0.05).The expression levels of Nrf2/NRE signaling pathway related proteins(Nrf2,Keap1,HO-1,NQO1)in lung tissue of model group were significantly lower than those in the normal group(P<0.05,P<0.01).Compared with the model group,the expression levels of Nrf2,Keap1,HO-1,NQO1 in the Qingfei Tongluo Recipe middle and high dose groups were significantly increased(P<0.05),while that of Qingfei Tongluo Recipe group was significantly increased(P<0.05).The expression of Nrf2,Keap1,HO-1 and NQO1 in the Qingfei Tongluo Recipe high dose group was significantly increased than that in the low dose group(P<0.05);the expression level of Keap1 in the Qingfei Tongluo Recipe high dose group was significantly higher than that in the medium dose group(P<0.05).Conclusion:Qingfei Tongluo Recipe can protect the
