针刺对干眼兔结膜胆碱能抗炎通路的影响 被引量:7

Effects of acupuncture on the cholinergic anti-inflammatory pathway in the conjunctiva of dry eye rabbits

摘要 目的:通过对干眼模型兔结膜相关胆碱能抗炎通路与特异性生物学指标分析,探讨针刺治疗干眼眼表炎症的作用机制。方法:36只健康新西兰兔随机分为空白组、模型组、假针刺组、针刺组、西药组、针药结合组,每组6只。空白组每日皮下注射0.9%氯化钠溶液,其余各组每日4次皮下注射氢溴酸东莨菪碱每次2.0 mg/kg,均连续35d。造模第22天开始,西药组双眼滴氟米龙滴眼液;针刺组电针治疗,穴位为睛明、丝竹空、攒竹、瞳子髎、太阳穴,双侧取穴,每日1次,留针15 min;假针刺组用钝头针点刺治疗;针药结合组用氟米龙滴眼液和电针治疗同时进行治疗。检测各组泪液分泌量(SⅠT)、泪膜破裂时间(BUT)、角膜荧光染色(CFS),光镜观察结膜组织结构,采用酶联免疫法检测结膜乙酰胆碱(ACh)、α7烟碱型乙酰胆碱受体(α7nAChR)和白介素-6(IL-6)含量,并应用Western Blot法检测结膜内p-STAT3、JAK2蛋白的表达情况。结果:治疗第35天,与模型组比较,针刺组、西药组和针药结合组SⅠT、BUT和CFS均显著改善(P<0.05);针刺可减少结膜上皮细胞的脱落,改善淋巴细胞浸润状态;同时针刺组、西药组、针药结合组结膜ACh、α7nAChR含量均显著升高(P<0.05),而结膜IL-6含量显著降低(P<0.05);结膜组织中JAK2、p-STAT3蛋白表达量显著降低(P<0.05)。结论:针刺能够调节干眼模型兔SⅠT、BUT和CFS情况,增加结膜中α7nAChR和ACh的含量,降低IL-6的含量,同时调节结膜组织中JAK2/p-STAT3蛋白的表达量。 Objective:To explore the mechanism of acupuncture in the treatment of dry eye surface inflammation by analyzing the conjunctival related cholinergic anti-inflammatory pathway and specific biological indicators in dry eye model rabbits.Methods:Thirty-six healthy New Zealand rabbits were randomly divided into six groups:blank group,model group,sham-acupuncture group,acupuncture group,western medicine group and acupuncture+western medicine group.In the blank group,saline was injected subcutaneously every day for 35 days.In the other groups,scopolamine hydrobromide was injected subcutaneously 4 times a day for 35 days,2.0 mg/kg each time.From the 22 nd day,the western medicine group was treated with fluorometholone eye drops,the acupuncture group was treated with electroacupuncture,the points were Jingming(BL 1),Cuanzhu(BL 2),Sizhukong(TE 23),Tongziliao(GB 1)and Taiyang(EX HN5),15 minutes/time/day;the sham acupuncture group was treated with blunt needle;the acupuncture+western medicine group was treated with fluorometholone eye drops and electroacupuncture at the same time.Schirmer I test(SIT),tear break time(BUT),corneal fluorescence staining(CFS)were measured.And the conjunctival tissue structure was observed under light microscope.The contents of ACh,α7-nicotinic acetylcholine receptor(α7 nAChR)and IL-6 in conjunctiva were detected by enzyme-linked immunosorbent assay.The expression of p-STAT3 and JAK2 in conjunctiva was detected by Western Blot.Results:After 35 days treatment,compared with the model group,SIT,BUT and CFS in the acupuncture group,western medicine group and acupuncture+western medicine group were significantly improved(P<0.05).Acupuncture could reduce the detachment of conjunctival epithelial cells and improve the infiltration of lymphocytes;compared with the model group,ACh andα7 nAChR in the conjunctiva of the acupuncture group,western medicine group and acupuncture+western medicine group were significantly increased(P<0.05),IL-6 and the expression of JAK2,p-STAT3 protein in the acupuncture gr
